人参皂苷Rg3对子宫内膜细胞生长及凋亡的影响

摘要 目的比较人参皂苷Rg3干预前后3种子宫内膜细胞的生长、增殖与凋亡状况,探讨人参皂苷Rg3抑制子宫内膜异位症(EMs)的作用机制。方法流式细胞术测不同浓度Rg3对子宫内膜细胞的周期及凋亡的影响。结果 (1)中低浓度人参皂苷Rg3可以对3种子宫内膜细胞均产生抑制作用,抑制率与对照组比较差异有统计学意义(P<0.01);随着与药物孵育时间的延长,药物浓度增加,细胞增殖明显抑制。(2)中低浓度人参皂苷Rg3即可促进3种子宫内膜细胞凋亡活性。(3)中、高浓度人参皂苷Rg3使3种内膜细胞发生G1期阻滞,G0/G1期细胞增加,S期细胞减少,有效抑制子宫内膜细胞有丝分裂前期蛋白质和ATP的合成,使子宫内膜细胞增殖速度减慢。结论人参皂苷Rg3具有抑制3种子宫内膜细胞增殖的作用。其作用机制可能是阻滞细胞的G0/G1期和促进细胞的凋亡活性。 Objective To compare the growth, proliferation and apoptosis of three Upes of endometrial cells before and after Ginsenosicle Rg3 intervention, and to explore the mechanism of Ginsenosicle Rg3 in inhibiting ectopic enclometriosis. Methods Flow cytometry was used to measure the effects of different concentrations of Rg3 on the cell cycle and apoptosis of enclometrial cells. Results ① Low and meclium concentrations of Ginsenosicle Rg3 were found to inhibit all the three Upes of enclometfial cells with the inhibition rates being significantly different from that in the control group (P〈0.01). The inhibition of cell proliferation was more prominent along with longer incubation time and higher Rg3 concentration. ② Low and meclium concentrations of Ginsenosicle Rg3 promoted the apoptotic activity of three Upes of enclometrial cells. ③ Meclium to high concentration of Ginsenosicle Rg3 caused Gi phase arrest, increased number of Go/Gi-phase cells, and fewer number of S-phase cells in three types of enclometrial cells, thereby effectively inhibiting the synthesis of proteins early inmitosis and ATP in enclometrial cells, and uhimately slowing clown the proliferation of enclometrial cells. Conclusion Ginsenosicle Rg3 may inhibit the proliferation of three types of enclometrial cells. The unclerlying mechanism may be thearrest of G0/G1 cell phase and promotion of apoptotic activity of these cells.
