摘要
目的氨基胍干预糖尿病大鼠,探讨在糖尿病大鼠阴茎中终末糖基化产物(AGEs)和胱硫醚-1-裂解酶(CSE)及硫化氢(H2s)的关系。观察在糖尿病勃起功能障碍中AGEs对H2S通路的影响。方法6周龄清洁级雄性Wister大鼠,体重180-220g,60只,随机分A组(10只)、B组(10只)和模型组(40只)。A、B组自由饮水,饮食;模型组自由饮水,高脂高糖饮食1个月造糖尿病模型,成模23只,随机分成C组12只,D组11只。A组继续自由饮水,饮食;B组正常饮食,饮水中按1g/L加入氨基胍;C组高脂高糖饮食,自由饮水;D组高脂高糖饮食,饮水中按1g/L加入氨基胍。6周后测量大鼠阴茎海绵体内压(ICP)和平均颈动脉压(MAP),血清、阴茎组织H2S浓度,应用免疫组织化学和免疫印记观察阴茎组织中AGEs、终末糖基化产物受体(RAGEs)、CSE的表达。结果比较A组(0.64±0.03)与B组(0.65±0.03)ICP/MAP、H2S含量(P=0.123);比较C组(0.40±0.01)与D组(0.44±0.02)ICP/MAP(P=0.001);阴茎中AGEs、RAGEs、CSE和H2S含量差异有统计学意义;A组与C组ICP/MAP,阴茎中AGEs、RAGEs、CSE和H2S含量差异有统计学意义。结论在糖尿病大鼠中AGEs通过RAGEs介导影响CSE和H2S生成,进而影响大鼠勃起功能。
Objective Diabetic rats were treated with aminoguanidine, in order to investigate the relationship between advanced glyeation end products (AGEs), eystanthionine -γ-lyase (CSE) and hy- drogen sulfide (H2S) in diabetic rats penis. It was to confirm that AGEs impacts the erectile of the rats with diabetes mellitus erectile dysfunction through hydregen sulfide pathway. Methods The cleaning grade Wister rats of 6 weeks old, male, weighing about 180 -220 g, a total of 60 rats, were randomly di- vided into group A (10 rats), group B( 10 rats) and model group(40 rats). Group A and group B with free drinking and eating. The modal group were given free drinking water, high fat and high - glucose diet for 1 month. In model group, 23 diabetic model rats were induced, which were randomly divided into group C of 12 rats and group D of 11 rats. Group A were continued to feed with free drinking and eating, group B were continued to normal diet and every litre drinking water was added 1 gram aminoguanidine; group Cwere continued to feed with high fat and high - glucose diet, free drinking water; group D were continued to high fat and high - glucose diet, every litre drinking water was added 1 gram aminoguanidine. After 6 weeks of feeding, each group rats were measured the intracaveruous pressure and mean arterial pressure. The concentration of hydrogen sulfide in serum and penis tissue were measured. Meanwhile AGEs, receptor for advanced glycation end products (RAGEs), CSE in penile tissue were observed through immunohisto- chemical ; RAGEs and CSE in penile tissue were measured by Western blotting. Results The difference of ICP/MAP and the concentration of hydrogen sulfide between the group A and the group B which were trea- ted by aminoguanidine had no statistical significance (P=0. 125 ) ; the difference of intracavernous pres- sure (ICP)/mean arterial pressure (MAP) between the group C and the group B was statistical signifi- cance, and the difference of contents of AGEs, RAGEs, CSE and H2S in the penis between the group C and the group B was statistical significance ( P=0. 001 ) ; the difference of ICP/MAP and contents of AGEs, RAGEs, CSE and H2S in the penis between the group A and the group C was statistical signifi- cance. Conclusion In diabetes mellitus rats, AGEs can affect the production of CSE and H2S through RAGEs, and then affect erectile function in the rats.
