脂肪干细胞联合筋膜干细胞修复大鼠骨性关节炎 被引量:2

Adipose derived stem cells in combination with Fascial derived stem cells for the treatment of osteo-arthritis in rat

摘要 目的探讨联合使用脂肪间充质干细胞和筋膜间充质干细胞治疗大鼠骨性关节炎的效果。方法从大鼠腹股沟处脂肪垫分离脂肪间充质干细胞、后肢骨骼肌筋膜分离筋膜间充质干细胞,经鉴定后扩增培养,两种细胞按不同比例混合,分为A组(1:0)、B组(5:1)、C组(1:1)、D组(1:5)、E组(0:1),F(O:O)组,总数为1×10。,注射进由碘乙酸诱导的大鼠骨性关节炎关节腔内。干细胞注射治疗12周后获取各组大鼠后肢膝关节标本,分别进行大体和病理组织学评估。结果分离的脂肪间充质干细胞、筋膜间充质干细胞CD29、CD71、CD90阳性表达,CD45阴性表达;均可以进行多向诱导分化;当两种细胞按1:1混合治疗时大体观得分:A组(4.25±0.96)、B组(4.83±1.11)、D组(4.41±0.90)、E组(4.91±0.79)、F组(5.75±1.21)均高于C组(2.92±0.79,P=0.010)、病理组织学评分A组(8.16±1.46)、B组(7.41±1.78)、D组(8.08±1.44)、E组(8.83±1.47)、F组(15.58±2.77)均高于C组(5.83±1.85,P=0.010)。结论脂肪间充质干细胞联合筋膜间充质干细胞关节混合比例为1:1时,软骨修复效果最佳。 Objective To evaluate the effect of using Adipose derived mesenchymal stem cells (ADSCs) and Fascial derived mesenchymal stem cells (FDSCs) in the treatment of osteoarthritis in rats. Methods Isolated from rat groin fat pad between adipose mesenchymal stem cells, hind legs between skeletal muscle membrane separation membrane, mesenchymal stem cells were identified after amplification culture, The two ceils were mixed in different proportions and divided into groups A (1:0), B (5:1), C ( 1 : 1 ), D ( 1:5 ), E ( 0:1 ) and F ( 0:0 ), a total of 1 × 10^6, injected by iodine osteoarthritis joint cavity of rats induced by acetic acid. After 12 weeks of stem cell injection treatment, the posterior limb knee joint samples of each group of rats were obtained for gross and pathological histological evaluation. Results The positive expressions of CD29, CD71 and CD90 of fat mesenchymal stem cells and fascial mesenchymal stem cells were detected, and the negative expressions of CD45 were detected. All of them can be multidirec- tional induced differentiation. When the two cells were mixed by 1:1, the gross score was:Group A (4.25±0.96), groupB (4.83±1.11), groupD (4.41±0.90), groupE (4.91±0.79), groupF ( 5.75±1.21 ) were all higher than group C ( 2. 92±0.79, P = 0. 010 ) , The pathological histological score of group A (8. 16 ±1.46), groapB (7.41±1.78), groupD (8.08±1.44), groupE (8.83±1.47 ) and group F ( 15.58±2. 77) were all higher than group C (5.83 ± 1.85, P = O. 010). Conclusion When the ratio of fat mesenchymal stem cells and fascia mesenchymal stem cells is 1 : 1, the best effect of cartilage renair is achieved.
