白藜芦醇对一次性力竭运动大鼠心肌NLRP3炎性小体表达的影响 被引量:6

Effects of Resveratrol on the Expression of Myocardial NLRP3 Inflammasome in Rats After A Single Bout of Exhaustive Exercise

摘要 目的:探讨白藜芦醇对一次性力竭运动大鼠心肌NLRP3炎性小体表达的影响及机制。方法:3月龄雄性SD大鼠30只,随机分为对照组(C)、一次性力竭运动组(E)和白藜芦醇+一次性力竭运动组(RE),每组10只。RE组腹腔注射白藜芦醇,注射剂量为5 mg/kg体重,每天1次,连续7 d;其余2组注射同等剂量生理盐水。E组和RE组在末次注射24 h后进行一次性力竭运动。训练结束后即刻腹腔麻醉,血流动力学方法评定心功能,Western blot检测心肌SIRT1、HMGB1、NLRP3、ASC-1、caspase-1和IL-1β蛋白表达,RT-q PCR检测心肌SIRT1 mRNA表达。结果:与C组比较,E组大鼠LVEDP明显升高(P<0.01),LVSP、±dp/dt_(max)和心率均显著下降(P<0.01),心肌SIRT1蛋白和mRNA表达显著减少(P<0.01),HMGB1、NLRP3、ASC-1、caspase-1和IL-1β蛋白表达均显著增加(P<0.01)。与E组比较,RE组大鼠LV-EDP明显下降(P<0.01),LVSP、±dp/dt_(max)和心率均明显上升(分别为P<0.01,P<0.05,P<0.01),心肌SIRT1蛋白和mRNA表达显著增多(P<0.01),HMGB1、NLRP3、ASC-1、caspase-1和IL-1β蛋白表达均显著减少(P<0.01)。结论:白藜芦醇可通过增加心肌SIRT1表达,抑制HMGB1表达,降低NLRP3炎性小体表达,缓解一次性力竭运动所致炎性应激状态,提升心功能。 Objective:This study explored the effect of resveratrol on the expression of myocardial NLRP3 inflammasome in rats after a single bout of exhaustive exercise.Methods:Thirty male Sprague Dawley rats were randomly divided into control group(C),a single bout of exhaustive exercise(E)and a single bout of exhaustive exercise with resveratrol group(RE)(n=10).Rats in RE group were injected intraperitoneally with resveratrol at a dose of 5 mg/kg body weight,once a day for 7 days.The other groups were given the same dose of normal saline.Rats in groups E and RE were performed an acute exhaustive treadmill exercise 24 h after the last injection.The abdominal anesthesia was operated immediately after the end of training,cardiac function was tested by hemodynamic method,the protein expressions of myocardial SIRT1,HMGB1,NLRP3,ASC-1,caspase-1 and IL-1βwere examined by Western blotting,and the mRNA expression of myocardial SIRT1 was tested by RT-q PCR.Results:Compared with C group,LVEDP and protein expressions of myocardial HMGB1,NLRP3,ASC-1,caspase-1 and IL-1βwere significantly increased in the E group(P0.01);LVSP,±dp/dt_(max),heart rate and the protein and mRNA expression of myocardial SIRT1 were significantly decreased in E group.Compared with E group,LVEDP and protein expressions of myocardial HMGB1,NLRP3,ASC-1,caspase-1 and IL-1βwere significantly decreased in the RE group(P〈0.01);LVSP,±dp/dt_(max),heart rate and the protein and mRNA expressions of myocardial SIRT1 were significantly increased in RE group.Conclusions:Resveratrol alleviated the inflammatory stress induced by a single bout of exhaustive exercise and improved cardiac function by increasing the expression of SIRT1 and decreasing the expression of HMGB1and NLRP3 inflammasome.
