花生红衣下调Bax、Bak表达防治化疗后血小板减少症的研究

Study on peanut red skin in treating chemotherapy-induced thrombocytopenia by downregulating the expression of Bax, Bak protein

目的:研究花生红衣对化疗后血小板减少症的小鼠外周血小板(PLT)、脾脏指数及Bax、Bak蛋白表达的影响。方法:采用腹腔注射环磷酰胺法建立化疗后血小板减少症的小鼠模型。随机分为模型组,花生红衣低、中、高剂量组(简称花低、中、高组),rh IL-11组,空白组,每组14只。花低、中、高组(0.25、0.50、1.00g/mL,0.1mL/20g)灌服花生红衣;空白组和模型组灌服等量蒸馏水;rh IL-11组皮下注射0.25μg/g rh IL-11,每日1次,共7d。给药7d后,检测小鼠外周PLT、脾脏指数。采用Western Blot方法检测Bax、Bak的蛋白表达。结果:治疗7d后,花低、中、高组PLT和脾脏指数明显高于模型组(P<0.01)。花生红衣可下调Bax、Bak蛋白表达。结论:花生红衣可能通过下调Bax、Bak蛋白表达而抑制PLT凋亡,提高小鼠免疫力,从而达到治疗化疗所致的血小板减少症。

Objective： To investigate the effect of peanut red on peripheral blood platelet, spleen index and Bax, Bak protein expression in mice with thrombocytopenia after chemotherapy. Methods： The model of chemotherapy-induced thrombocytopenia was successfully established by peritoneal injection of cyclophosphamide. The mice randomly divided into 6 groups： model group, low dose group with peanut red skin, middle dose group with peanut red skin, high dose group with peanut red skin, rh IL-11 group and control group, 14 mice each group. Control group and model group were given the same amount of distilled water; rh IL-11 group was given the 0.25μg/g of rh IL-11 by subcutaneous injection; Peanut red skin group was given low, medium and high dose of peanut red skin. All groups were administrated once a day for 7 days. After 7 d administration, peripheral blood platelet（PLT） and spleen index were detected. Protein expression of Bax and Bak was detected by Western Blot. Results： After 7 d treatment, the PLT count of the low, medium and high dose peanut red group was significantly higher than that of the model group, and the spleen index was significantly higher than that of the model group（P〈0.01）. Peanut red could down-regulate Bax and Bak protein expression. Conclusion： Peanut red color may improve the immunity of mice by inhibiting the apoptosis of PLT, so as to cure the thrombocytopenia caused by chemotherapy.
