温通活血乳膏对糖尿病周围神经病变大鼠VEGF及细胞凋亡的影响

Effects of Wentong Huoxue Cream on VEGF and apoptosis indiabetic peripheral neuropathy rats

目的:探讨温通活血乳膏在糖尿病周围神经病变(DPN)大鼠中对血管内皮生长因子(VEGF)及细胞凋亡的影响。方法:建立链脲佐菌素及高糖高脂饲料诱导的DPN大鼠模型,随机分为:空白组、模型组、二甲双胍组、辣椒碱+二甲双胍组、温通活血乳膏+二甲双胍组。药物干预4周,观察大鼠一般情况,光镜下检测坐骨神经病理结构,免疫组化法检测VEGF的表达,并用TUNEL法检测背根神经节神经元细胞凋亡情况。结果:与模型组相比,温通活血乳膏+二甲双胍组的坐骨神经病理结构趋于正常,VEGF表达呈阴性;且神经细胞凋亡数目减少(P<0.01,P<0.05)。结论:温通活血乳膏抑制了DPN大鼠坐骨神经结构的改变,并抑制了VEGF的表达,阻止细胞凋亡的进程,从而延缓周围神经病变进展。

Objection： To investigate the effect of Wentong Huoxue Cream on VEGF and apoptosis in diabetic peripheral neuropathy rats. Methods： The DPN rat model induced by streptozotocin and high glucose and high fat diet was establish. Rats were randomly divided into blank group, model group, metformin group, capsaicin plus metformin group, Wentong Huoxue Cream plus metformin group. Drug intervention for 4 weeks, the general situation of rats was observed, the pathology of sciatic nerve was detected under the light microscopy; the expression of VEGF was examed by immunohistochemistry; the neuronal apoptosis in dorsal root ganglion neurons was detected by TUNEL method. Results： Compared with the model group, the sciatic nerve pathology tends to normal and the VEGF expression was negative, while the number of neuronal apoptosis was reduced in Wentong Huoxue Cream plus metformin group than that in the model group （P〈0.01, P〈0.05）. Conclusion： Wentong Huoxue Cream could inhibit the change of sciatic nerve structure in DPN rats, and inhibit the expression of VEGF and prevent the process of apoptosis, thus delaying the progression of peripheral neuropathy.
