Analysis on Nucleosides in Cordyceps militaris from Different Sources

摘要 采用超高效液相色谱法测定了不同来源蛹虫草样品中的9种核苷类成分,包括尿嘧啶、尿苷、肌苷、鸟苷、腺嘌呤、胸苷、腺苷、2’-脱氧腺苷、虫草素,使用统计产品与服务解决方案(19.0)和中药色谱指纹图谱相似度评价系统(2012)软件,分析不同来源蛹虫草核苷类成分的含量差异。结果表明,不同来源蛹虫草9种核苷类成分总含量在4 488.36μg·g^(-1)~10 294.16μg·g^(-1),平均值为8 115.06μg·g^(-1),变异系数为15.33%;蛹虫草中9种核苷类成分含量高低依次为腺苷、鸟苷、尿苷、虫草素、肌苷、腺嘌呤、尿嘧啶、 2’-脱氧腺苷、胸苷,其中,腺苷、鸟苷、尿苷和虫草素是蛹虫草中主要的核苷类成分;聚类分析结果显示,不同来源蛹虫草中核苷类成分的含量具有一定的地理相关性。指纹图谱相似度计算结果显示, 30个样品间指纹图谱相似度在0.69~0.99,表明不同来源蛹虫草核苷类成分相似度较高,但仍然存在一定的差异。 A Ultra Performance Liquid Chromatography (UPLC) method has been used for the simultaneous determination of 9 nucleosides (uracil, uridine, inosine, guanosine, thymidine, adenine, adenosine, 2'-deoxyadenosine and cordycepin) in Cordyceps militaris, Statistical Product and Service Solutions (19.0) and the Similarity Evaluation System for Chromatographic Fingerprint of TCM (2012) were used to analyze the content of nucleosides in C. militaris from different sources. The total amount of the 9 nucleosides was between 4 488.36 μg·g^-1 and 10 294.16 μg·g^-1, the average of the 9 analytes was 8 115.06 μg·g^-1, and the coefficient of variation (CV) was 15.33%. The content of the 9 nucleosides in C. militaris from high to low in turn was: adenosine, guanosine, uridine, cordycepin, inosine, adenine, uracil, 2'-deoxyadenosine and thymidine, and the adenosine, guanosine, uridine, cordycepin were the main nucleosides in C. militaris. The cluster analysis showed that the content of nucleosides correlated to their producing areas to a certain extent. The similarity of UHPLC fingerprints of nucleosides among 30 samples was between 0.69 and 0.99, which indicated that the nucleosides in C. militaris from different sources were of high similarity, however, there was some difference among samples.
