摘要
目的探究"智三针"对高强度训练致大鼠肝脏损伤的防治作用。方法将6~8周SD大鼠随机分为正常组、模型组和治疗组。正常组常规喂养;模型组每天以高强度运动60 min,每周6次,持续8周;治疗组除高强度运动外,每天给予"智三针"治疗1次。观察各组大鼠一般生存状态、体质量;考马斯亮兰法检测肝脏中蛋白含量,比色法测定肝脏中肝糖原含量,黄嘌呤氧化酶法测定肝脏中超氧化物歧化酶(SOD)活性,TBA法测定肝脏中丙二醛(MDA)含量;光镜下观察肝脏的组织病理学变化,透射电镜检查肝脏的超微结构变化。结果与模型组比较,治疗组大鼠的一般生存状态有所改善,体质量均有不同程度增加;治疗组大鼠蛋白含量、肝糖原水平、肝脏SOD均明显升高(P<0.05,P<0.01),MDA水平显著降低(P<0.01)。病理和电镜结果显示,治疗组大鼠肝细胞损伤均得到一定程度改善。结论 "智三针"可改善长期高强度训练致大鼠肝脏损伤,对肝脏损伤具有一定保护作用,其主要机制可能与改善大鼠肝脏过氧化损伤有关。
Objective To investigate the preventive and therapeutic effects of "Zhi San Zhen" acupuncture on liver injury induced by high intensity training in rats. Methods SD rats aged 6-8 weeks were randomly divided imo the normal group, model group and treatment group. The normal group was treated with routine feeding. The model group was treated with high intensity exercise at intensity of 60 minutes everyday, once a day, six times a week for 8 weeks. Based on the high intensity exercise, the treatment group was treated with "Zhi San Zhen" acupuncture once a day. The general survival state and body weight of rats in each group were observed. The protein content in the liver was detected by Coomassie blue staining, the liver glycogen content in the liver was detected by colorimetric assay, the superoxide disluutase(SOD) activity in the liver was determined by xanthine oxidase method, and the MDA content in the liver was detected by TBA method. The histopathological changes of liver were observed under light microscope, and the ultrastructural changes of liver were examined by transmission electron microscopy. Results Compared with the model group, the general survival state of rats was improved in the treatment group and the body weight was increased in varying degrees. The protein content, fiver glycogen teve| and liver SOD activity were obviously increased in the treatment group ( P〈O.05, P〈O.OI ) , and the MDA level was significantly decreased (P〈0.01). The results of pathology and electron microscopy showed that the damage of hepatocyte was improved in the treatment group, Conclusion "Zhi San Zhen" acupuncture can alleviate the liver injury caused by long-term high intensity training in rats and exert the protective effects on liver injury. The main mechanism may be related to the improvement on liver peroxidation injury in rats.
