抗衰老酶1在心脏病中的作用及相关药物对其调控作用的研究进展 被引量:1

摘要 抗衰老酶1(Sirt1)是细胞中广泛存在的一种去乙酰化酶,在心血管疾病中发挥重要作用。近年来研究表明,Sirt1可增强细胞抗氧化应激的能力,减少细胞凋亡,减轻炎症反应,调节细胞能量代谢,从而减小心肌缺血再灌注后的梗死面积,促进心脏功能恢复;Sirt1缺失可导致扩张型心肌病,但对心肌肥大的作用则受外源刺激类型、Sirt1表达量变化程度等因素影响;心力衰竭会影响心肌细胞中Sirt1的表达和定位,Sirt1失活可造成心力衰竭;激活Sirt1可减少多柔比星、放射等刺激造成的心脏损伤;白藜芦醇、雌激素、褪黑素及部分中草药提取物对心脏的保护作用也需要Sirt1参与。本文对Sirt1在心脏病及相关药物调控中的作用研究进行综述,有助于推动Sirt1作为药物治疗靶点进入临床,探寻更有效的心脏病治疗方法。 Sirtuin1(Sirt1),one protein deacetylase,plays a vital role in heart diseases.Recent studies have indicated that Sirt1 could reduce myocardial infarct size and promote the recovery of heart function after ischemia-reperfusion by enhancing resistance to oxidative stress,inhibiting apoptosis,attenuating inflammatory responses,and regulating energy metabolism of cells.Sirt1 deficiency could lead to dilated cardiomyopathy.However,its role in myocardial hypertrophy is influenced by the type of exogenous stimuli and the magnitude of Sirt1 expression.Heart failure could affect the expression and localization of Sirt1 in cardiac myocytes,and inhibiting the activity of Sirt1 may cause heart failure.Activating Sirt1 could alleviate myocardial injury induced by doxorubicin,irradiation and other stimuli.In addition,the protective effect of resveratrol,estrogen,melatonin and some Chinese herbal medicine extracts on heart diseases also require Sirt1 participation.The role of Sirt1 in heart diseases and related drug regulation reviewed in this article will contribute to the promotion of Sirt1 as a therapeutic target and to the exploration of more effective treatment of heart diseases.
