青薯10号茎尖脱毒及快繁技术研究

摘要 以马铃薯品种青薯10号为试材,研究不同消毒方法、不同浓度激素配比的诱导培养基、不同茎尖大小、不同培养条件对茎尖脱毒和成苗效果的影响。结果表明:自来水冲洗1~2min,0.05%次氯酸钠溶液浸泡10min后用无菌水冲洗3~5次,可达到理想的消毒效果;茎尖分生组织0.3~0.4 mm(带2个叶原基)、诱导培养基:MS+1.0 mg/l BA+0.05 mg/l NAA+30g/l蔗糖+7g/l琼脂、光照时长14 h/d、光照强度2000lx条件下培养温度28℃时成苗效果最好。 Potato Qingshu No.10 as experimental materials, study of different disinfection methods, concentration of different ratio of hormone induced medium, different stem tip size, different culture conditions on the stem tip detoxication and sprout effect. The results showed that the ideal disinfection effect could be achieved by washing water for 1-2min and 0.05% NaClO for 10min with sterile water for 3-5 times. Stem tip meristem 0.3- 0.4 mm (2 leaf primordium), the induction culture medium:MS + 1.0mg/l BA+0.05 mg/l NAA + 30 g/l sucrose + 7 g/l AGAR, illumination 14 h/d, light intensity under the condition of 2000 lx sprout best temperature 28℃.
