FTY720对耐顺铂非小细胞肺癌细胞增殖及凋亡的影响

Study on effect of FTY720 on proliferation and apoptosis of cisplatin-resistance of non-small cell lung cancer cell

目的:检测芬戈莫德(fingolimod,FTY720)对耐顺铂非小细胞肺癌细胞株A549增殖及凋亡的影响。方法:应用CCK-8检测耐顺铂A549细胞株在FTY720不同浓度(5μmol/L、10μmol/L、15μmol/L和20μmol/L)作用不同时间(24 h和48 h)后的增殖活性。流式细胞仪检测不同浓度(5μmol/L、10μmol/L和20μmol/L)作用不同时间(24 h和48 h)后耐顺铂A549细胞凋亡情况,Western blot法检测细胞凋亡相关蛋白的表达。结果:耐顺铂A549细胞的增殖抑制程度和促进凋亡的程度均与FTY720呈时间依赖及剂量依赖性。结论:FTY720以时间依赖及剂量依赖模式对耐顺铂A549细胞呈现出抑增殖及促凋亡作用。

Objective ：To investigate the effect of FTY720 on proliferation and apoptosis of cisplatin - resistance of non - small cell lung cancer cell. Methods：CCK-8 assay and flow eytometry were introduced to detect the effect of different acting dose and different acting time FTY720 on proliferation and apoptosis of eisplatin-resistance A549. Western blot analysis was used to test the level of apoptosis-relative protien. Results ：With the increasing of FTY720 concentration and acting time, the proliferation inhibiton and apoptosis promotion of FTY720 in A549/DDP cell were enhanced. Conclusion：FTY720 could inhibit A549/DDP cell growth in time-and dose-dependent manner in vitro.
