RhoA在胃癌中的异常表达及意义

Abnormal expression and significance of RhoA in gastric carcinoma

目的检测胃癌中Rho A的表达程度,分析其与胃癌患者临床病理因素的关系,并探讨其在胃癌发生发展过程中的生物学意义。方法收集78例胃癌病例、24例癌旁组织石蜡包埋标本,以18例正常胃黏膜组织标本为对照组,利用免疫组化方法,检测其中Rho A的表达情况,分析不同组织Rho A蛋白表达的差异及胃癌组织Rho A与患者临床病理学特征的关系。结果 Rho A在胃癌、癌旁和正常胃黏膜组织中的阳性表达率分别是59.0%,25.0%和22.2%,Rho A蛋白在胃癌组织中的表达比癌旁组织或正常胃黏膜组织的表达高,且差异具有统计学意义(P<0.05)。Rho A蛋白表达与性别、年龄无关(P>0.05),与肿瘤浸润深度、淋巴结转移和分化程度有关(P<0.05)。结论 Rho A蛋白表达异常可能与胃癌的发生发展有关,参与了胃癌的浸润转移,可作为胃癌侵袭、转移的一个重要生物学标志。

Objective To investigate the expression of RhoA in gastric carcinoma and its correlation with clinicopathological features, and explore its biological significance in the occurrence and development of gastric carcinoma.Methods A total of 78 cases of gastric carcinoma and 24 cases of pericarcinomatous tissues were collected in form of paraffin embedding samples, and 18 specimens of normal gastric mucosal tissue samples were collected as controls. The expression of RhoA was detected by immunohistochemistry. The difference of RhoA protein expression in different tissues and its correlation with clinicopathological features were analyzed.Results The positive rates of RhoA were 59.0%,25.0% and 22.2% in gastric carcinoma, adjacent tissues and normal mucosa, respectively. The expression of RhoA was higher in gastric carcinoma than that of adjacent tissues or normal gastric mucosal tissues（ P 〈0.05）. RhoA protein expression was not related to sex and age of patients（ P 〉0.05）,but it was correlated to tumor invasion stages, lymph node metastasis and degree of differentiation（ P 〈0.05）.Conclusion The expression of RhoA protein may be related to the occurrence and development of gastric carcinoma, and it is involved in the invasion and metastasis of gastric carcinoma, which can be regarded as an important biological marker for the invasion and metastasis of gastric carcinoma.
