摘要
目的酶法优化女贞子多糖提取工艺。方法利用纤维素酶处理女贞子提取多糖,分别以酶解时间、酶解温度、酶量和p H值为主要影响因素,采用L_9(3~4)正交实验设计进行提取工艺的优化。结果通过正交试验及验证性实验确定其最佳提取工艺条件为酶解时间120 min,酶解温度40℃,酶量1%,p H值为5.2。在此条件下,女贞子多糖提取率可达6.58%。结论酶法提取的女贞子多糖具有显著的抗氧化活性,对DPPH自由基、羟自由基和超氧阴离子自由基作用明显,是一种良好的天然抗氧化剂。
Objective To optimize the extraction of polysaccharides from Fructus ligustri lucidi by enzyme method. Methods The polysaccharides were extracted from Fructus ligustri lucidi with cellulase,and the effect of enzymolysis time,enzymolysis temperature,cellulase concentration and pH value were chosen as influence factor.Extraction conditions were optimized by L 9(3^ 4)orthogonal experiment. Results The results showed that optimum technical parameters through orthogonal test and verifying experiments were:enzymolysis time of 120 min,enzymolysis temperature of 40 ℃ ,cellulase concentration of 1%,and pH value of 5.2.Under above conditions,the extraction ratio of polysaccharides from Fructus ligustri lucidi was 6.58%. Conclusion The polysaccharides from Fructus ligustri lucidi with cellulase treatment is considered as a nature antioxidant which possess relatively higher antioxidant acitivity and scavenging activities on DPPH,hydroxyl radical and superoxide anion radicals.
