桑叶1-脱氧野尻霉素水提液对高脂饮食小鼠脂肪分解代谢的影响 被引量:11

EFFECT OF 1-DEOXYNOJIRIMYCIN WATER EXTRACT FROM MULBERRY LEAVES ON LIPID CATABOLISM IN HIGH-FAT FED MICE

摘要 目的讨论了桑叶1-脱氧野尻霉素(1-deoxynojirimycin,DNJ)水提液对高脂血症的预防作用,同时初步探索其对脂肪分解途径的影响。方法用高脂饲料饲喂小鼠的同时,灌胃不同剂量桑叶DNJ水提液,4w之后,采集小鼠血液和肝脏,用试剂盒测定血清中各项生化指标的水平,及血液、肝脏中脂肪分解关键酶的活性。结果与高脂对照组相比,高剂量桑叶DNJ水提液能使小鼠体重增加率降低16.40%(P<0.05),腹腔脂肪系数下降23.62%(P<0.05),肝脏指数下降7.25%(P<0.05);使血液总胆固醇(total cholesterol, TC)、总甘油三酯(total triglyceride, TG)、低密度脂蛋白(low-density lipoprotein, LDL)含量分别下降42.66%、16.91%、22.29%(P<0.05),高密度脂蛋白(high-density lipoprotein,HDL)含量增加40.03%(P<0.05);使血液中天门冬氨酸转移酶(aspartate aminotransferase, AST)和丙氨酸转移酶(alanine aminotransferase, ALT)活力分别下降28.72%、28.23%(P<0.05);使血液中激素敏感性脂肪酶(hormone sensitive lipase,HSL)、肝脂酶(hepatic lipase, HL)活性分别上升18.62%、16.98%(P<0.05),肝脏中脂蛋白脂酶(lipoprotein lipase, LPL)、HL活性分别上升15.49%(P<0.05)、7.82%。结论适量桑叶DNJ水提液可以增加脂肪分解代谢酶的活性,加速脂肪动员,促进脂肪消耗。 Objective To explore the effect of 1-deoxynojirimycin (DNJ) water extract from mulberry leaves on hyperlipidemia and lipid catabolism pathway. Method Mice were not only fed with a high-fat diet,but also treated with DNJ water extraction at different doses. After 4 weeks, we collected blood and liver samples to determine the biochemical parameters, and the activity of related enzymes. Results Compared with high-fat group, the highest dosage of DNJ water extraction decreased weight-gain by 16.40%, abdominal fat coefficient by 23.62%, liver index by 7.25%, contents of TC, TG, LDL in serum by 42.66%, 16.91% and 22.29% respectively, and increased content of serum HDL by 40.03%, activities of AST and ALT in serum by 28.72% and 28.23% respectively, activities of HSL and HL in the liver by 18.62% and 16.98% respectively, activities of LPL and HL in the liver by 15.49% and 7.82% respectively. Conclusion A suitable amount of DNJ water extraction from mulberry leaves can accelerate fat metabolism and promote fat utilization by increasing the activity of related enzymes in lipid catabolism.
