复方烫伤膏对深Ⅱ度烧伤模型大鼠创面修复及Smad2、Smad3、CTGF表达的影响 被引量:3

摘要 目的:探究复方烫伤膏对深Ⅱ度烧伤模型大鼠创面修复及Smad2、Smad3、结缔组织生长因子(CTGF)表达的影响。方法:将大鼠分为正常对照组、模型组、蜂花烧烫伤膏组、复方烫伤膏低剂量组、复方烫伤膏中剂量组、复方烫伤膏高剂量组;ELISA检测大鼠血清中白细胞介素6(IL-6)和肿瘤坏死因子(TNF-α)的含量,检测大鼠烫伤组织含水量;检测烫伤组织微血管密度(MVD);masson染色检测烧伤组织形态变化;蛋白免疫印迹(WB)检测Smad2、Smad3、p-Smad2、p-Smad3、CTGF表达情况。结果:复方烫伤膏组大鼠血清中IL-6、TNF-α含量以及烧伤组织含水量显著低于模型组(P<0.05);复方烫伤膏高剂量组大鼠血清中IL-6、TNF-α含量以及烧伤组织含水量与蜂花烧烫伤膏组相比,差异无统计学意义(P>0.05);复方烫伤膏组大鼠烧伤组织MVD及p-Smad2、p-Smad3、CTGF表达水平显著低于模型组(P<0.05);复方烫伤膏高剂量组大鼠烧伤组织MVD及p-Smad2、p-Smad3、CTGF表达水平与蜂花烧烫伤膏组相比,差异无统计学意义(P>0.05)。结论:复方烫伤膏有助于深Ⅱ度烧伤模型大鼠创面修复,推测这一作用是通过调控Smad2、Smad3、CTGF的表达水平实现的,为深Ⅱ度烧伤的临床治疗提供一定的理论依据。 Objective:To explore the effect of Compound Scald Ointment on the wound repair and the expressions of Smad2,Smad3 and connective tissue growth factor(CTGF) in rats with deep partial thickness burn.Methods:The rats were divided into normal controlled group,model group,Melissa Burn Ointment group,Compound Scald Ointment low dose group and Compound Scald Ointment middle dose group,Compound Scald Ointment high dose group;The contents of serum interleukin 6(IL-6) and tumor necrosis factor(TNF-α) in rats was detected by ELISA,the water content of the burn tissue in the rats was measured;The microvascular density(MVD) of the burn tissue was detected;Masson staining was used to detect the morphological changes of burn tissue;The expressions of Smad2,Smad3,p-Smad2,p-Smad3 and CTGF was detected by protein immunoblotting(WB).Results:The contents of serum IL-6,TNF-α and water content of burn tissue in the rats of the Compound Scald Ointment group were significantly lower than those in the model group(P〈0.05);Compared the contents of serum IL-6,TNF-αand water content of burn tissue in the rats of the Compound Scald Ointment high dose group and the Melissa Burn Ointment,the difference was not statistically significant(P〉0.05);The expression levels of MVD,p-Smad2,p-Smad3 and CTGF in the burn tissue of the Compound Scald Ointment group was significantly lower than those in the model group(P〈0.05);Compared the expression levels of MVD,p-Smad2,p-Smad3 and CTGF in the burn tissue of the Compound Scald Ointment high dose group and the Melissa Burn Ointment,the difference was not statistically significant(P〉0.05).Conclusion:Compound Scald Ointment is helpful for wound healing of deep partial thickness burn rats,it is speculated that this effect is achieved by regulating the expression levels of Smad2,Smad3 and CTGF,this providing a theoretical basis for the clinical treatment of deep partial thickness burn.
