犬新孢子虫NC-GFP株在4种细胞中培养的比较 被引量:3

摘要 为筛选出犬新孢子虫的最佳培养细胞,本试验对利用非洲绿猴肾细胞(Vero)、人胚肾细胞(HEK-293)、人宫颈癌细胞(HeLa)和小鼠骨髓瘤细胞(SP2/0)培养犬新孢子虫NC-GFP株进行了比较研究,连续培养犬新孢子虫15d,观察Vero细胞、HEK-293细胞、HeLa细胞及SP2/0细胞的形态变化并计数犬新孢子虫的数量,绘制虫体在细胞中生长曲线。结果表明,Vero细胞、HEK-293细胞、HeLa细胞及SP2/0细胞均可培养犬新孢子虫,尤其在HEK-293细胞和Vero细胞中虫体生长较快,分别在接种虫体的第5,6天虫体数量达到最大值;而犬新孢子虫在SP2/0细胞和HeLa细胞中生长缓慢,分别在第7,8天数量达到最大值,数量处于较低水平。综合比较,HEK-293细胞可以短期较快的培养犬新孢子虫,可作为一种新的细胞系培养犬新孢子虫。本试验为体外细胞培养犬新孢子虫提供了科学依据。 In order to screen out the best for the culture of Neospora canium,in this experiment,we observed and compared the Neospora caninum cultivated with the African green monkey kidney cell line,the human embryonic kidney cell line,the human cervical carcinoma cell line and mouse SP2/0 myeloma cell.Continuously cultivated Neospora caninumfor 15 days,observed the morphological change of Vero cells,HEK-293 cells,HeLa cells and SP2/0 cells and counted the number of larvae.Then,drawing the growth curve of the larvae in the cell.The results showed that these four cell lines all could cultivate the Neospora caninum,especially in HEK-293 cells and Vero cells,the larvae grew faster,with the maximum number of larvae at 5 th and 6 th days after inoculating larvae.However the Neospora grew slowly in HeLa and SP2/0 myeloma cells,and the number of parasites reached a maximum at the 7 th and 8 th days,the number was at a low level.After careful comparison,HEK-293 could be used to cultivate Neospora in a short time and was used as a new cell line to culture Neospora caninum.This experiment provided a scientific basis for the in vitro culture Neospora caninum.
