产前冷应激对妊娠母鼠肝脏炎症及细胞凋亡的影响 被引量:2

Effects of prenatal cold stress on the liver inflammation and apoptosis of rats in pregnancy

摘要 为探讨产前冷应激对妊娠期大鼠肝脏的影响。本研究选择20只体质量(230±20)g的健康SPF级雌性Wistar大鼠,预饲1周,合笼交配。受孕后随即分为常温对照组(RT)和冷应激组(C)2组,每组10只。RT组全程处于(22±2)℃、相对湿度(40.0±0.1)%的环境中饲养,C组在妊娠第15天转移到(4.0±0.1)℃,相对湿度(40.0±0.1)%环境中进行冷刺激。在妊娠第17,21天,各组随机选取5只进行麻醉处死,采集肝脏组织,用Western blot的方法检测肝脏中IL-1β、Bcl-xL和Bax的蛋白表达量。结果显示,在妊娠第17天(冷刺激3d),C组大鼠肝脏中IL-1β的表达量显著高于RT组(P〈0.05);肝脏中Bcl-xL/Bax比值显著降低(P〈0.05)。结果表明冷应激可能会导致妊娠期大鼠肝脏发生炎症反应,并促进细胞凋亡,对肝脏产生一定损伤。 In order to discuss the effects of prenatal cold stress on the liver of rats in pregnancy.Select 20 healthy SPF female wistar rats,who weigh 230 to 250 grams,and give a week to mate with male wistar rat.After conception,they were divided into normal temperature control group(RT group)and cold stress group(C group)and 10 of each group.The RT group was kept in an environment of about 22 degrees and 40 percent humidity.Group C moved to about 4 degrees on the15 th day of gestation,with 40 percent humidity.At the 17 th and 21 st days of gestation,each group randomly selected 5 rats to be executed by anesthesia,collected liver tissue.And used the western blot method to measure the IL-1β,Bcl-xL and the Bax expression of protein in the liver.The results showed that at the 17 th day of gestation(the cold stimulus 3 days),the expression of IL-1βin the liver of rats with cold stress was higher than that of normal environment group(P〈0.05);the BclxL/Bax ratio in the liver of cold stress was significantly reduced in 3 days(P〈0.05).The results showed that the cold stress could cause inflammation in the liver of the pregnant rats and promote apoptosis,resulting in some certain damage to the liver.
