自噬相关蛋白p62表达与乳腺癌预后关系Meta分析

Meta-analysis of the expression of p62 protein and survival in breast cancer

目的探讨乳腺癌组织中自噬相关蛋白p62表达与乳腺癌预后之间的关系。方法检索Pub Med、The Cochrane Library和Web of Science数据库,检索时限均从建库至2018年1月,收集p62表达与乳腺癌预后关系的相关研究。主要结局指标包含总存活期(OS)和无病存活期(DFS);以风险比为效应量,各效应量以95%可信区间表示,采用STATA 12. 0统计学软件进行Meta分析。结果本研究共纳入符合标准的文献6篇,总样本1 653例。Meta分析结果显示,p62阳性表达乳腺癌患者发生死亡的风险是p62阴性表达患者的2. 54倍(风险比=2. 54,95%可信区间1. 63~3. 98,P <0. 05)。p62阳性表达乳腺癌患者复发和转移的风险是p62阴性表达患者的2. 32倍(风险比=2. 32,95%可信区间1. 72~3. 13,P <0. 05)。结论 p62在乳腺癌组织中高表达的患者预后差,可以作为判断乳腺癌转移、侵袭能力的预后指标,可能成为乳腺癌基因治疗的新靶点。

Objective To explore the relationship of p62 with the breast cancer survival by Meta-analysis.Methods Search PubMed,The Cochrane Library and Web of Science databases,the retrieval time is from the database construction to January 2018,the correlation between p62 expression and prognosis of breast cancer was collected.Main outcome measures include total survival （OS） and disease-free survival （DFS）;Taking the risk ratio as the effect size,each effect size was expressed in a 95% confidence interval,and Meta analysis was performed using STATA 12.0 statistical software.Results A total of 6 literatures meeting the standards were included in this study,with a total of 1653 samples.Meta analysis showed that the risk of death in patients with breast cancer with positive p62 expression was 2.54 times higher than that in patients with negative p62 expression （ HR =2.54,95% CI ：1.63～3.98, P 〈0.05）.The risk of recurrence and metastasis of breast cancer patients with positive p62 expression was 2.32 times higher than that of patients with negative p62 expression （ HR =2.32,95% CI ：1.72～3.13, P 〈0.05）.Conclusion The prognosis of patients with high expression of p62 is poor in breast cancer.It can be used as a prognostic marker to predict metastasis and invasion ability and may become a new target for gene therapy of breast cancer.
