右美托咪定对CLP小鼠肺损伤不同时间Nrf2表达的影响

Effects of Dexmedetomidine on Expression of Nrf2 at Different Times after Lung Injury in CLP Mice

目的观察右美托咪定预处理对盲肠结扎穿孔(CLP)脓毒症模型小鼠急性肺损伤的不同时间点Nrf2表达的影响。方法 48只小鼠随机分为三组,假手术(SHAM)组,模型(CLP)组和右美托咪定(DEX)组,每组16只,各组在造模前30min分别腹腔注射等剂量生理盐水或右美托咪定100μg·kg-1,随后行CLP造模并于术后8h和24h应用荧光定量PCR测定肺组织Nrf2 m RNA表达,Western Blot测定Nrf2蛋白表达,HE染色观察肺组织病理结构变化。结果与CLP组比较,DEX组肺组织的损伤情况明显减轻,Nrf2 m RNA及Nrf2蛋白表达亦显著增加(P <0. 01)。结论右美托咪定预处理能减轻脓毒症引起的24h内肺组织损伤,这种作用可能与上调Nrf2有关。

To observe the effects of dexmedetomidine pretreatment on the expression of Nrf2 at different time points in acute lung injury in cecaligation and puncture （CLP） sepsis model mice.METHODS Forty-eight mice were randomly divided into three groups：sham-operated （SHAM） group,model CLP） group,and dexmedetomidine （DEX） group,16 in each group.Each group was given intraperitoneal injection of equal doses of normal saline or dexmedetomidine（100μg·kg^ -1 ） 30 min before modeling,then followed by establishing CLP model.8h and 24h after operation,the expression of Nrf2 mRNA in lung tissue was detected by fluorescence quantitative PCR,Nrf2 protein expression was measured by Western Blot,HE staining was used to observe the pathological changes of lung tissue.RESULTS Compared with the CLP group,the damage of the lung tissue in the DEX group was significantly reduced,and the expression of Nrf2 mRNA and Nrf2 protein was also significantly increased（ P 〈0.01）.CONCLUSIONS The retreatment of dexmedetomidine can reduce the lung tissue damage caused by sepsis within 24 hours,this effect may be related to upregulation of Nrf2.
