美洲大蠊提取物对肝硬化门静脉高压大鼠门静脉压力及巨噬细胞移动抑制因子表达的影响 被引量:1

摘要 目的探讨美洲大蠊提取物(PAE)对门静脉高压大鼠门静脉压力及肝组织巨噬细胞移动抑制因子(MIF)表达的影响。方法将60只SD大鼠按随机数字表法分为正常对照组、模型组、PAE高剂量组(PAE1组)、PAE中剂量组(PAE2组)、PAE低剂量组(PAE3组)、心得安组,每组10只。除正常对照组外,其余各组采用四氯化碳综合法造模。自造模日起,正常对照组、模型组给予生理盐水灌胃,PAE1组、PAE2组、PAE3组按0.5、0.25、0.10 g/kg的剂量给予PAE灌胃,心得安组按0.18 mg/100 g的剂量给予普奈络尔灌胃,共8周。观察各组大鼠一般情况,干预结束后比较各组大鼠门静脉压力、肝组织的MIF蛋白及mRNA表达水平。结果模型组大鼠一般情况差于正常对照组,门静脉压力高于正常对照组,但各药物干预组均低于模型组,心得安组低于PAE2组、PAE3组(均P<0.05);PAE1组、PAE2组MIF蛋白及mRNA表达水平均低于模型组、PAE3组及心得安组(均P<0.05)。结论 PAE可以降低门静脉高压大鼠的门静脉压力,这可能与其降低MIF水平有关。 Objective To investigate the effects of Periplaneta americana extract (PAE) on the portal pressure and expression of liver tissue macrophage migration inhibitory factor( MIF) in the cirrhotic rats with portal hypertension. Methods Sixty SD rats were divided into normal control group, model group, high-dose PAE group( PAE1 group), medium-dose PAE group (PAE2 group ), low-dose PAE group (PAE3 group) ,and propranolol group according to the random number table method,with 10 rats in each group. Except the normal control group,the other groups used the CCL4 comprehensive method for modeling. From the day of modeling, the normal control group and model group were given normal saline by gavage,the PAE1 group ,PAE2 group and PAE3 group received PAE of 0.5,0.25 and 0. 10 g/kg a day by garage respectively,and the propranolol group was given propranolol of 0.18 mg/100 g a day by gavage. All groups were intervened for 8 weeks. The general condition of rats were observed in each group,and the portal pressure and the expression levels of liver tissue MIF protein and mRNA were compared among six groups after intervention. Results The general condition of rats in the model group was poorer than that in the normal control group; the portal pressure of rats in the model group was higher than that in the normal control group ,but the portal pressure in the drug intervention groups were lower than that in the model group, and the portal pressure in the propranolol group was lower than that in the PAE2 group or PAE3 group(all P 〈0.05) ;the expression levels of MIF protein and mRNA in the PAE1 group and PAE2 group were lower than those in the model group, PAE3 group and propranolol group( all P 〈 0.05 ). Conclusion PAE can reduce the portal pressure of cirrhotic rats with portal hypertension,which may be related to its effects on reducing MIF level.
