摘要
[目的]探讨双酚A对小鼠脾细胞中树突状细胞(DC)构成比的影响.[方法]取成年BALB/c雌性小鼠,按照体质量随机分为对照组和4个双酚A剂量(0.1,1.0,10.0,100.0mg/kg)组,以灌胃的方式染毒双酚A,1周3次,连续5周,利用流式细胞仪测定脾细胞数目及脾细胞中DC的百分比.[结果]10.0,100.0mg/kg的双酚A显著增加脾细胞数目(P<0.01),且显著增高浆细胞样树突状细胞(CD11clowmPDCA1+细胞)的百分比和数目(P<0.05,P<0.01);1.0,10.0,100.0mg/kg的双酚A显著增高髓样树突状细胞(CD11chigh MHCII+细胞)的百分比和数目(P<0.05,P<0.01).[结论]双酚A增加脾细胞数目,改变脾细胞中DC的构成比.
OBJECTIVE To explore the effects of bisphenol A(BPA)on the percentage of dendritic cells(DC)in mouse spleen cells.METHODS Adult female BALB/c mice were selected.They were randomly divided into control group and 4 different dose of BPA groups(0.1,1.0,10.0 and 100.0 mg/kg)according to body mass.Mice were exposed to BPA by gavage,3 times per week for 5 weeks.The number of spleen cells and the percentage of DC in spleen cells were measured by flow cytometer.RESULTSBPA(10.0 and 100.0 mg/kg)significantly increased the number of spleen cells(P〈0.01),and increased the percentage and the number of plasmacytoid dendritic cells(CD11 clowmPDCA1+cells)in the spleen(P〈0.05,P〈0.01).BPA(1.0,10.0 and 100.0 mg/kg)evidently enhanced the percentage of myeloid dendritic cells(CD11 chigh MHCII+cells)in the spleen(P〈0.05,P〈0.01).CONCLUSION BPA could increase the number of spleen cells and change the composition ratio of DC in spleen cells.
