摘要
[目的]研究扛板归乙醇提取物(EPP)对大鼠离体血管功能的影响及其机制.[方法]制作大鼠离体胸主动脉血管环标本,观察EPP(0.1~100.0mg/L)对内皮完整和去内皮的大鼠离体胸动脉血管舒张作用的影响,并探讨作用机制.[结果]EPP剂量依赖性地舒张PE(1.0μmol/L)预收缩的内皮完整的血管环,而内皮去除、L-NAME和ODQ预处理可显著抑制EPP对血管环的舒张效应(P<0.05);维拉帕米、吲哚美辛、格列本脲及TEA预处理不能抑制EPP诱导的内皮完整血管环的舒张作用(P>0.05).[结论]EPP具有内皮依赖性血管舒张作用,其机制认为可能与NO-cGMP途径有关系.
OBJECTIVE To investigate the effects of ethanol extract of Persicaria perfoliata(EPP)on the function of isolated blood vessels in rats and its related mechanism.METHODSSpecimens of isolated rat thoracic aortic rings were prepared.Observe the vasodilation effects of EPP(0.1-100.0 mg/L)on isolated rat thoracic aorta with or without intact endothelium and explore its mechanism.RESULTS EPP dosedependently relaxed the thoracic aortic rings with intact endothelium which were contracted by phenylephrine(1.0μmol/L).Endothelium removal,pretreatment with L-NAME and ODQ could significantly inhibit the relaxation effects of EPP on aortic rings(P〈0.05).However the relaxation effects of EPP were not blocked by pretreatment with the verapamil,indomethacine, glibenclamide and tetraethylammonium(P〉0.05).CONCLUSION EPP had the function of endothelial-dependent vasodilatation.Its mechanism might be related with nitric oxide/cGMP signaling pathway.
