摘要
目的检测肝癌患者血清中miRNA的表达水平。方法提取肝癌患者血清中的miRNA进行芯片杂交测序,并用qRT-PCR的方法比较确定患者miRNA与健康对照的表达水平变化,检测手术前手术后肝癌患者血清中相关miRNA的表达水平变化。结果与健康对照组相比,肝癌患者血清中hsa-miR-206、hsa-miR-20a-3p、hsa-miR-411-5p、hsa-miR-122和has-miR-423水平明显升高,术后hsa-miR-206、hsa-miR-411-5p、hsa-miR-122和has-miR-423水平显著降低; hsa-miR-26a-5p、hsa-miR-455-5p、hsa-miR-30e-3p、hsa-let-7f-5p和hsa-miR-6852水平显著降低,术后hsa-miR-26a-5p和hsa-miR-455-5p水平显著回升。结论血清miRNA可作为肝癌诊断的依据,并能反映疾病发展过程。
Objective To detect the expression levels of serum miRNA in liver cancer patients. Methods Extract mitlNA from serum collected from liver cancer patients and send them for RNA-seq and microassay analysis. Quantatitive Real-Time PCR was used to evaluate the expression levels of candidate miRNAs for result comfirmation. In addition, expression profiles of candidate miR- NAs were assessed by qRT-PCR before and after surgery. Results Compared with healthy control, expression levels of hsa-miR-206, hsa-miR-20a-3p, hsa-miR-411-Sp, hsa-miR-122 and has-miR-423 in patient serum were increased sharply, and after surgery levels of hsa-miR-206, hsa-miR-411-5p, hsa-miR-122 and has-miR-423 decreased; the miRNA decreasing their expression levels in liver canc- er patients included hsa-miR-26a-5p, hsa-miR-455-Sp, hsa-miR-30e-3p, hsa-let-Tf-Sp and hsa-miR-6852, while after surgery hsa- miR-26a-5p and hsa-miR-455-5p increased their expression levels. Conclusions Certain serum miRNAs could be used as biomarkers for liver cancer diagnosis and reflect disease prognostic progress.
