二仙汤及其拆方对去卵巢大鼠骨质疏松症的影响

Effects of Erxian Decoction and Its Separate Prescriptions on Ovariectomy Induced-Osteoporosis in Rats

目的：探讨二仙汤及其拆方防治骨质疏松症的作用及初步机制,为临床用药加减提供理论依据。方法：实验采用去卵巢骨质疏松大鼠模型,以二仙汤整方为标准,根据君、臣、佐、使药进行拆分原方,在二仙汤君药基础上,共分为8组,即假手术组、模型组、福善美组（0.1 mg·kg-1·d-1）、二仙汤整方（A方）、淫羊藿＋仙茅（B方）、淫羊藿＋仙茅＋巴戟天（C方）、淫羊藿＋仙茅＋当归（D方）和淫羊藿＋仙茅＋知母＋黄柏（E方）,动物依据分组分别给药,连续90 d后,收集样本,分别检测骨密度、骨微细结构、骨生物力学及血清ACP5、OPG变化。结果：二仙汤及其拆方能增加骨密度,改善骨微细结构,增加骨小梁数量,增加股骨三点弯曲应力等,且能抑制血清ACP5水平,A方和E方的作用尤为明显。结论：二仙汤整方能有效防治骨质疏松,君药是防治骨质疏松的基础,配伍佐药知母、黄柏能明显增加骨量、防止骨微细结构破坏。二仙汤及其拆方防治骨质疏松的作用可能与抑制骨吸收有关。

Objective： To explore the effect of Erxian Decoction and its separate prescriptions on ovariectomy induced-osteoporosis in rats. Methods： The ovariectomy induced-osteoporosis were established in rats and the separate prescriptions were assured according to the Erxian Decoction and its rule of the Chief,Adjuvant,Assistant and Courier herbs. All rats were randomly divided into 8 groups： sham,control,Fosamax and A-E prescriptions groups. All rats were administered herbs according to the divided group. The sham and control groups were administered with normal saline. All of samples were collected after 90 days. And the bone mineral density,bone microstructure,bone biomechanics and serum ACP5,OPG were measured. Results： The bone mineral density,bone microstructure,the number of trabecular bone and the femoral three-point bending stress were increased by Erxian Decoction and its separate prescriptions,and the ACP5 serum level decreased. The effect of A prescription on ovariectomy induced-osteoporosis was equal to that of E prescription in rats. Conclusion： It is confirmed that the effect of Erxian Decoction on ovariectomy induced-osteoporosis in rats. The Chief herbs of Erxian Decoction are the basis of the effect of Erxian Decoction on ovariectomy induced-osteoporosis. The bone mineral mass,the bone microstructure and the bone biomechanics would be improved after the application of Adjuvant herbs,such as Rhizoma Anemarrhenae,Cortex Phellodendri. The effect of Erxian Decoction and its separate prescriptions on ovariectomy induced-osteoporosis may be relevant to inhibition of bone resorption.
