药对川芎-香附对硝酸甘油偏头痛大鼠单胺类神经递质、一氧化氮和一氧化氮合酶的影响 被引量:15

摘要 目的:分析川芎-香附药对治疗偏头痛的有效提取部位及其治疗偏头痛的作用机制。方法:大鼠随机分组,灌胃给药,1次/d,连续5 d,末次给药30 min后,皮下注射硝酸甘油10 mg·kg-1造模,检测血清中NO和NOS,脑干中NE、DA、5-HIAA、5-HT含量。结果:造模后大鼠5-HIAA、5-HT显著降低(P〈0.05),NO、NOS显著升高(P〈0.05),NE、DA降低却无统计学差异。阳性药组,川芎-香附水提物和醇提物组均可显著升高模型大鼠5-HT(P〈0.05),降低NO、NOS(P〈0.05),阳性药组和醇提物还可显著升高5-HIAA(P〈0.05)。川芎-香附醇提物高、中剂量组可显著升高模型大鼠5-HIAA、5-HT(P〈0.05),减低NO和NOS(P〈0.05)。结论:川芎-香附醇提物较水提物治疗偏头痛效果更优,川芎-香附可能通过调节神经递质和血管活性物质NE、DA、5-HT、5-HIAA、NO、NOS的释放,发挥治疗偏头痛的作用。 Objective: To explore the anti-migraine effective extract of drug pair of Chuanxiong Rhizoma and Cyperi Rhizoma,and explain the anti-migraine mechanism. Methods: All rats were randomly divided into different groups and administered intragastricly once each day for 5 days. 30 min after the last administration,all groups were injected subcutaneously nitroglycerin 10 mg·kg-1 to induce migraine models except normal control group. The contents of NO and NOS in serum and NE,DA,5-HIAA and 5-HT in the tissue of brain stem were determined. Results: Compared to normal rats,the levels of 5-HIAA and 5-HT in migraine rats deceased significantly( P 〈0. 05). NO and NOS increased significantly( P〈 0. 05),but NE and DA decreased without significant difference. The positive drug,ethanol extract and water extract could improve 5-HT and reduce NO and NOS in migraine rats( P 〈0. 05). The positive drug and ethanol extract could also enhance 5-HIAA in migraine rats( P〈 0. 05). The high and medium dose groups' ethanol extract could significantly improve 5-HIAA and 5-HT and reduce NO and NOS in migraine rats( P 〈0. 05). Conclusion: The ethanol extract was better than water extract in treating migraine. Chuanxiong Rhizoma and Cyperi Rhizoma could treat migraine by regulating the levels of 5-HT,5-HIAA,NO and NOS.
