健脾中药胃肠安对胃癌MKN45细胞侵袭迁移能力的影响

Effects of Herbs for Strengthening on Invasion and Migration Ability of Gastric Cancer MKN45 Cells

目的：研究健脾中药胃肠安对胃癌细胞MKN45侵袭、迁移等生物学行为的影响及其分子机制。方法：体外培养MKN45细胞,分为胃肠安方低剂量（0.5 mg·mL-1）组、高剂量（1 mg·mL-1）组和对照组,采用Transwell小室实验评价胃肠安方对胃癌细胞MKN45侵袭能力的影响、细胞划痕实验检测胃肠安对MKN45细胞迁移能力的影响,Western Blotting检测经胃肠安干预后的MKN45细胞中金属基质蛋白酶（MMPs）MMP2、MMP9的蛋白水平。结果：Transwell侵袭和迁移实验显示,胃肠安干预后的MKN45细胞穿膜数明显减少;细胞划痕实验显示,胃肠安可显著抑制MKN45细胞迁移;Western Blot结果显示,胃肠安干预后的MKN45细胞的MMP2、MMP9蛋白表达水平明显降低。结论：胃肠安可降低MKN45细胞的侵袭能力,抑制胃癌细胞迁移。胃肠安抑制人胃癌细胞MKN45的侵袭和迁移,其机制可能与下调MMP2、MMP9蛋白水平直接相关,这可能是胃肠安影响胃癌细胞侵袭与转移的机制之一。

Objective： To study the effect of traditional Chinese herbs for strenghtening spleen on the biological behavior of gastric cancer cell MKN45 invasion and migration and its molecular mechanism. Methods： MKN45 cells were cultured in vitro and divided into gastrointestinal Angola 0. 5 mg/mL,1 mg/m L groups and control group. Transwell assay was used to evaluate the influence of Weichangan Formula on gastrointestinal invasion activity of gastric cancer. The cell scratch assay was used to evaluate the influence on MKN45 migration ability. And We detected MMP2 and MMP9 protein levels in MKN45 cells after gastrointestinal surgery by Western blot. Results： Transwell invasion and migration experiments showed that the number of MKN45 cells was significantly decreased after gastrointestinal surgery and showed a concentration dependent. The cell scratch test showed Weichang＇an significantly inhibited MKN45 cell migration. Western blotting results showed that after Weichang＇an intervention for MKN45 cells,the expression levels of MMP2 and MMP9 protein decreased significantly. Conclusion： Weichang＇an reduces the invasive ability of MKN45 cells and inhibits cell migration. The inhibition of invasion and migration of human gastric cancer cell line MKN45,which may be related to the down regulation of MMP2 and MMP9 protein,may be one of the mechanisms of invasion and metastasis of gastric cancer cells.
