摘要
目的:建立柱前衍生化HPLC比较梅花鹿和马鹿茸多糖的单糖组成和含量的差异。方法:采用超声和水煎煮分别提取梅花鹿和马鹿茸多糖,经三氟乙酸水解后,用PMP试剂进行柱前衍生化并进行HPLC分析。结果:梅花鹿和马鹿茸多糖的单糖组成种类相同,均是由甘露糖、盐酸氨基葡萄糖、核糖、葡萄糖醛酸、半乳糖醛酸、盐酸氨基半乳糖、葡萄糖、半乳糖8种单糖组成,但组成比例不同,且梅花鹿茸多糖含量和得率高于马鹿茸。结论:建立了鹿茸多糖中单糖组成的高效液相色谱研究方法。
Objective: To establish a pre-column derivation HPLC method for the comparison of monosaccharide compositions and content difference between sika deer and red deer antler polysaccharides. Method: The polysaccharides of sika deer and red deer antler were extracted by ultrasonic and water decoction and hydrolyzed into monosaccharides with TFA. The hydrolysate was derivatized with PMP and analyzed via HPLC. Result: The species of monosaccharide composition between sika deer and red deer antler polysaccharide was the same,which was composed of mannose,Glucosaminehydrochloride,ribose,glucuronicacid,galacturonicacid,ammoniumgalactosehydrochloride,glucose,galactose,but the composition proportion was different. And sika deer antler polysaccharide content and yield higher than the red deer antler.Conclusion: A high performance liquid chromatography method for the determination of monosaccharides in the pilose antler polysaccharides was established.
