摘要
藻蓝蛋白是一种重要的天然功能性食品添加剂。本文以人类肺癌LTEP-a-2细胞为模型,采用流式细胞术、四甲基偶氮唑蓝(MTT)检测以及划痕愈合等试验,分别从细胞凋亡、细胞增殖和细胞迁移3个方面研究藻蓝蛋白对体外培养的肺癌LTEP-a-2细胞的影响,结果表明,藻蓝蛋白能引起细胞形态发生异常改变;通过上调促凋亡基因和下调抑凋亡基因的表达引起细胞的凋亡;通过异常调控细胞周期G1/S转换点相关基因的表达使细胞阻滞在G1期,同时,藻蓝蛋白通过调控迁移相关基因的表达抑制LTEP-a-2细胞的体外迁移能力。研究结果证实藻蓝蛋白对肺癌LTEP-a-2细胞的体外生长具有显著的抑制作用,这为新型抗肺癌功能食品的开发以及肺癌的治疗提供理论基础。
C-phycocyanin (C-PC) is an important natural functional food additive. In order to investigate the effect of C-PC on LTEP-a-2 ceils, we performed flow cytometry, MTT assay and wound healing experiments to explore apop- tosis, proliferation and migration of LTEP-a-2 cells. The results showed that C-PC could cause abnormal changes in cell morphology. It also induced apoptosis by up-regulating expression of pro-apoptotic genes and down-regulating the anti- apoptotic genes. C-PC could regulate the expression of G1/S checkpoint related genes and lead to G1 phase arrest. In addition, it could inhibit the migration of LTEP-a-2 cells in vitro by regulating the expression of migration-related genes. The results indicated that C-PC had a significantly inhibitory effect on the growth of lung cancer LTEP-a-2 cells in vitro, which provided a theoretical basis for the development of new anti-lung cancer functional foods.
