摘要
优化提取三亚海胆中氨基酸工艺试验条件。研究利用酸水解法处理海胆样品,超声波辅助水解,单因素变量法考察单因素变量,正交试验优化水解条件,启用高相液相色谱仪测其氨基酸含量。结果表明,海胆氨基酸水解的最佳条件是:水解时间22 h、水解温度105℃、盐酸用量10 m L、超声波水解时间30 min;18种被测氨基酸线性关系良好,相关系数R2均在0.999 8以上,仪器检出限在0.000 015 96~0.002 131 07μg/L之间,各氨基酸的RSD在0.15%~1.57%之间,试验样品A(低含量0.1 mg/m L标准液)的氨基酸的回收率在97.9%~103.3%之间,RSD小于1.05%;样品B(高含量1.0 mg/m L标准液)的氨基酸的回收率在99.7%~100.4%之间,RSD小于0.09%,氨基酸总含量为310.98 mg/g。该方法是一项新的试验尝试,试验结果令人满意。
It is to optimize the extraction process of amino acid from sea urchin experimental conditions. The amino acid content has been measured, handling sea cucumber samples are used by acid hydrolysis, with ultrasonic assisted hydrolysis,taking single factor variable method and orthogonal experiment design. The results show that the optimum conditions for hydrolysis of amino acids in sea urchin are as follows: the hydrolysis time is 22 h, the hydrolysis temperature is 105 ℃, the amount of hydrochloric acid is 10mL, and the ultrasonic hydrolysis time is 30 min; 18 kinds of amino acids are measured with good linear relationship, the correlation coefficient R2 is above 0.999 8. The limit of the instrument is between 0.000 015 96-0.002 131 07 g/L, and the RSD of each amino acid is between 0.15% and 1.57%. The recovery of amino acid of the experimental samples of A(low content 0.1 mg/mL standard solution) is 97.9%-103.3%, RSD is less than 1.05%; The recovery of amino acid of the sample B(high content 1.0 mg/mL standard solution) is 99.7%-100.4%, RSD is less than 0.09%,the total amino acid content is 310.98 mg/g. This method is a new experiment with satisfactory results.
