摘要
2017年,关岭自治县坡贡镇某牛场112头关岭牛普遍出现体型消瘦,精神萎靡等症状,经现场诊断为牛蜱病,感染率达100%。为准确鉴定寄生于关岭牛的牛蜱种属,为驱虫提供依据,采用通用引物TITS2-F1/TITS2-R1,扩增牛蜱ITS2基因。对PCR产物进行测序,比对分析,并构建系统进化树。结果显示,ITS2序列长约1 000bp,与微小牛蜱的同源性为99.4%。基于ITS2的种系发育分析表明,关岭牛蜱虫分离株(GLT)与GenBank数据库公布的微小牛蜱位于同一分支。结果表明,该牛场牛群感染蜱虫为牛微小牛蜱。通过注射虫退(伊维菌素),连续治疗35d,病牛得到康复。
In 2017,a total of 112 cattle in Pogong Town,Guanling County,suffered from symptoms such as body thinness and listlessness.After on-site diagnosis of burdock disease,the infection rate reached 100%.In order to accurately identify the species of Burdock parasitic in Guanling cattle and provide basis for insect repellent,universal primers TITS2-F1/TITS2-R1 were used to amplify ITS2 gene of calf.The PCR products were sequenced,compared and analyzed,and a phylogenetic tree was constructed.The results showed that the ITS2 sequence was about 1000 bp in length and 99.4% homologous to the burdock.Germline development analysis based on ITS2 showed that the Guanting burdock mite isolate(GLT)was located on the same branch as the tiny burdock published in the GenBank database.The results showed that the cattle population of the cattle farm was infected with micro-burdock cattle.After 35 days of continuous treatment with insect retreat(Ivermectin),the diseased cattle recovered.
