摘要
为探索柑橘黄龙病的病因,获得柑橘黄龙病拮抗菌,从茂名市信宜市的几个乡镇柑橘园采取柑橘叶样品,通过平板培养的方式分离纯化柑橘叶片的内生细菌,并通过PCR扩增技术对其鉴定分析。结果:从所采的柑橘叶片上分离出来4株内生细菌,经过革兰氏染色鉴定以及显微观察,发现分离出的4株均为革兰氏阳性菌。随后成功对柑橘内生细菌的DNA进行提取,并进行PCR扩增、测序,确定为其内生菌种,该研究可用于柑橘黄龙病的检测分析。
In order to find the cause of citrus huanglongbing and access the citrus huanglongbing antagonistic bacteria, in this study, citrus leaf samples were collected from citrus groves of several townships in Xinyi, Maoming, and the endophytic bacteria of citrus leaves were isolated and purified by plate culture. The identification and analysis of the endophytic bacteria were carried out by PCR amplification. Results: Four endophytic bacteria were isolated from the citrus leaves which collected from citrus groves, after preliminary Gram staining identification and microscopic observation, they were found to be Gram-positive bacterias. Subsequently, the DNA of endophytic bacteria in citrus was successfully extracted, amplified by PCR and sequenced. The endophytic bacteria were identified, which laid the foundation for the etiology of citrus huanglongbing.
