摘要
目的探讨手术室空气中分离的大肠埃希菌和肺炎克雷伯菌产Amp C酶的情况,并对Amp C酶耐药表型与基因型进行分析。方法菌株鉴定采用Phoemix100微生物分析仪鉴定;产Amp C酶初选采用头孢西丁纸片扩散法;采用多重PCR和DNA测序检测分析Amp C酶基因型。结果 67株大肠埃希菌和肺炎克雷伯菌经头孢西丁敏感试验筛选出疑产Amp C酶菌株11株,并经PCR检测有9株菌株均在405 bp处出现阳性条带,再经DNA测序均为DHA-1型Amp C酶。结论手术室空气中分离的大肠埃希菌和肺炎克雷伯菌产Amp C酶检出率较高,流行基因型以DHA-1型为主。
Objective To investigate the Amp C enzyme produced by Escherichia coli and Klebsiella pneumonia isolated in the air of operating room and analyze the resistance phenotype and genotype. Methods Strain was identified by the Phoemix100 microbial analyzer. Enzeme producting Amp C was primarily screened by cefoxitin disk diffusion method. And Amp C enzyme genotypes were analyzed by multiplex polymerase chain reaction( PCR) and DNA sequencing. Results Eleven strains producing Amp C enzemy were screened from 67 strains of Escherichia coli and Klebsiella pneumoniae by cefoxitin susceptibility test.Nine strains were detected by PCR at 405 bp positive bands,and then diagnozed as DHA-1 type Amp C enzyme by DNA sequence. Conclusion The detection rate of Amp C gene in Escherichia coli and Klebsiella pneumoniae isolated from operating room air was higher,and the prevalence of genotype was DHA-1.
