一株浒苔拉乌尔菌HT15-8的菌种鉴定及其抗肿瘤活性初步研究

摘要 目的:鉴定浒苔共生微生物HT15-8的菌种,同时对其代谢产物进行初步的分离纯化和抗肿瘤活性研究。方法:发酵培养10株浒苔共生微生物,用细胞毒活性检测法分别对其代谢产物进行抗肿瘤活性的筛选。选取活性最好的菌株HT15-8进行形态观察、生理生化实验鉴定、16S rRNA基因序列同源性分析。利用倒置显微镜观察HT15-8代谢产物的活性组分对BEL-7402、A549细胞形态的影响。结果:菌株HT15-8属于拉乌尔菌属,倒置显微镜下观察发现,菌株HT15-8代谢产物的活性组分能够明显改变肿瘤细胞形态并最终死亡。结论:浒苔共生微生物HT15-8为拉乌尔菌属,其产生的代谢产物对BEL-7402和A549两种肿瘤细胞具有初步的抗肿瘤活性。 Objective: To identify the species of the symbiotic micro-organism HT15-8 of Brassica oleracea, and to isolate and purify its metabolites. Methods: Ten symbiotic microorganisms of Brassica oleracea were fermented and cultured, and their metabolites were screened for antitumor activity by cytotoxicity assay. The best active strain HT15-8 was selected for morphological observation, physiological and biochemical experimental identification, and 16 S r RNA gene sequence homology analysis. The effect of the active components of HT15-8 metabolites on the morphology of BEL-7402 and A549 cells was observed using an inverted microscope. Results: The strain HT15-8 belonged to the genus Raulia. It was observed under an inverted microscope that the active components of the metabolite HT15-8 significantly changed the morphology of the tumor cells and eventually died. Conclusion: The symbiotic microorganism HT15-8 of Brassica oleracea is a genus Raulia, and its metabolites have a preliminary anti-tumor activity on BEL-7402 and A549 tumor cells.
