“肾脑相济”电针疗法对帕金森病模型小鼠中脑黑质胶质细胞的影响 被引量:10

Effects of Electro-Acupuncture Therapy with Coordination between Kidney and Brain on Substantia Nigra Neuroglia Cell in Parkinson's Disease Model Mice

摘要 目的探讨电针治疗帕金森病的可能作用机制。方法将48只小鼠随机分为空白组、模型组、电针组和美多芭组,每组12只。除空白组外,其余各组采用1-甲基-4-苯基1236四氢吡啶(MPTP)腹腔注射建立帕金森病小鼠模型。空白组灌服生理盐水0.2 ml,美多芭组灌服美多芭混悬液50 mg/kg,每天1次。电针组给予"肾脑相济"电针疗法,针刺"肾俞""百会""太溪"穴,每日1次,每次20 min,每日取左侧或右侧交替针刺治疗。各组均干预4周。观察小鼠的行为状态,小鼠中脑黑质中酪氨酸羟化酶(TH)、胶质纤维酸性蛋白(GFAP)、离子钙接头分子(Iba-1)阳性细胞数及蛋白表达。结果模型组小鼠行为状态较空白组明显变差,美多芭组与电针组小鼠行为状态较模型组改善。与空白组比较,模型组小鼠中脑黑质TH阳性细胞数量及蛋白表达明显减少,GFAP和Iba-1阳性细胞数量及蛋白表达明显增多(P<0.05);与模型组比较,电针组和美多芭组小鼠TH阳性细胞数量及蛋白表达明显增多,GFAP和Iba-1阳性细胞数量及蛋白表达明显减少(P<0.05)。美多芭组与电针组各指标比较差异均无统计学意义(P>0.05)。结论 "肾脑相济"电针疗法抑制星形胶质细胞和小胶质细胞的活性,从而增加其对多巴胺能神经元的保护作用,可能是其治疗帕金森病的作用机制之一。 Objective To study the possible mechanism of electro-acupuncture therapy on Parkinson' s disease( PD). Methods A total of 48 mice were randomly divided into blank group,mode group,electro-acupuncture group and Meddoba group,with 12 mice in each group. 1-Methyl-4-phenyl 1, 2, 3, 6 tetrahydropyridine( MPTP) was used to establish PD model mice by intraperitoneal injection in all groups except blank group. 0. 2 ml saline solution was given to the blank group. Meddoba suspension( 50 mg/kg) was given to Meddoba group,once a day. Electroacupuncture stimulation with coordination between kidney and brain was used in electro-acupuncture group on"Shenshu"( BL23),"Baihui"( GV20) and"Taixi"( K13),once a day,and 20 minutes each time with unilateral alternative treatment. The interventions in each group were all continuous for 4 weeks. The behaviors,amount of positive cells and expression of tyrosine hydroxylase( TH),glial fibrillary acidic protein( GFAP),Ionized calcium bindingadaptor molecule-1( Iba-1) in substantia nigra of the mice were observed. Results The behaviors of mice in model group were significantly inferior to the blank group,while Meddoba group and electro-acupuncture group were better than model group. The amount of positive cells and expression of TH in substantia nigra was significantly reduced in the model group,while GFAP and Iba-1 were obviously increased( P〈0. 05). Compared with the model group,the amount of positive cells and expression of TH was significantly increased in electro-acupuncture group and meddoba group,while GFAP and Iba-1 were significantly reduced( P〈0. 05). The index in meddoba group and electro-acupuncture group had no significant difference( P〉0. 05). Conclusion Electro-acupuncture therapy with coordination between kidney and brain can inhibit the activity of astrocyte and microglia,and further potentiate the protection of dopaminergic neuron,which might be the mechanism of PD.
