滋阴潜阳方对2K1C高血压大鼠肾损伤的影响

Effect of Ziyin Qianyang Recipe on Renal Injury in 2KIC Hypertensive Rats

目的评估滋阴潜阳方对两肾一夹(2K1C)高血压大鼠肾损伤的干预作用影响。方法复制32只2K1C高血压大鼠模型,造模后2周评估造模情况,选取造模成功的24只大鼠随机分为滋阴潜阳组、贝那普利组、模型组,另取8只大鼠为假手术组。各组大鼠连续灌胃给药8周,分别检测造模2周、灌胃4周、灌胃8周大鼠尾动脉收缩压。末次给药取24 h尿微量白蛋白,次日处理大鼠,腹主动脉取血检测肌酐(SCR)、尿素氮(BUN),HE染色观察肾脏病理学变化。结果滋阴潜阳方能抑制大鼠血压升高(P<0.05);显著降低大鼠尿微量白蛋白浓度(P<0.05);SCR和BUN含量显著降低(P<0.05)。肾脏病理切片示滋阴潜阳组肾脏血管内皮、过度增生肥大的系膜细胞减小;系膜区体密度及细胞外基质明显减小;基底膜厚度接近正常。结论滋阴潜阳方对改善高血压大鼠肾损伤具有一定疗效。

Objective To evaluate the intervention effect of Ziyin Qianyang recipe on renal injury in 2 K1 C hypertensive rats. Methods Twenty-two 2 K1 C hypertensive rat models were duplicated and the model establishment was evaluated 2 weeks after the model was established. Twenty-four successful rats were randomly divided into Ziyinqianyang group,benazepril group and model group. 8 rats were selected as sham operation group. The rats in each group were given gavage for 8 weeks. The rats were respectively given 2 weeks of gavage,4 weeks of gavage and 8 weeks of gavage. Urine microalbumin was taken 24 hours after the last administration,and the rats were treated the next day. Serum creatinine（ SCR） and blood urea nitrogen（ BUN） were taken from the abdominal aorta and the pathological changes of kidney were observed by HE staining. Results Ziyin Qianyang recipe could inhibit the increase of blood pressure（ P 0. 05）,decrease the level of urine microalbumin（ P 0. 05） and decrease the contents of SCR and BUN in rats（ P 0. 05）. Kidney pathological sections showed that the renal vascular endothelium and renal hyperplasia and hypertrophy of mesangial cells decreased; the body density and extracellular matrix of mesangial area decreased significantly; the thickness of basilar membrane was close to normal. Conclusion Ziyin Qianyang recipe can relieve renal injury in hypertensive rats.
