脑髓康通过调控氧化应激及单胺类神经递质改善VCI大鼠情景记忆能力 被引量:9

Naosuikang Decoction Restores Episodic Memory Ability in VCI Rats via Regulating Oxidative Stress and Monoamine Neurotransmitters

摘要 目的探讨脑髓康改善血管性认知障碍(VCI)大鼠情景记忆能力的机理。方法应用结扎双侧颈总动脉造成大鼠脑区慢性灌注不足的方法制备VCI大鼠模型,将造模成功大鼠48只随机分为模型组,脑髓康低(0.1 g·kg^(-1))、中(0.2 g·kg^(-1))、高(0.4 g·kg^(-1))剂量组,各12只,另设假手术组12只,给予各组大鼠相应药液灌服4周,应用新物体识别试验检测各组大鼠情景记忆能力的变化,并测定大鼠脑中氧化应激相关指标丙二醛(MDA)、超氧化物歧化酶(SOD)及谷胱甘肽过氧化物酶(GSH-Px)含量的变化,测定大鼠脑中去甲肾上腺素(NE)、多巴胺(DA)及5-羟色胺(5-HT)等单胺类神经递质含量的变化。结果给予脑髓康治疗后,能够显著改善VCI大鼠的情景记忆能力(P<0.01),并且显著提高VCI大鼠脑中氧化应激相关指标SOD(P<0.01)及GSH-Px(P<0.01)的含量,此外,脑髓康还对VCI大鼠脑中单胺类神经递质NE(P<0.05)、DA(P<0.01)及5-HT(P<0.01)有显著的调控作用。结论新物体识别试验结果提示中药复方脑髓康能够改善VCI大鼠的情景记忆能力。相关生化指标检测提示脑髓康可能通过抑制VCI大鼠脑中氧化应激以及调节其脑中单胺类神经递质的水平,达到改善VCI大鼠情景记忆能力的作用。 Objective To elucidate the mechanisms of Naosuikang decoction on episodic memory ability improving in vascular cognitive impairment(VCI)rats.Methods The VCI rat models were prepared using bilateral common carotid artery occlusion method.Rats were given the corresponding drug solutions gavage daily.After 4 weeks,novel object recognition test was carried out to evaluate the episodic memory ability in rats.The levels of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and malonaldehyde(MDA)in brain tissues of rats were detected.The levels of norepinephrine(NE),dopamine(DA)and 5-hydroxytryptamine(5-HT)in brain tissues of rats were also detected.Results Naosuikang decoction could improve the episodic memory ability in VCI rats(P〈0.01).In addition,the levels of SOD(P〈0.01)and GSH-Px(P〈0.01)in brain tissues of VCI rats were increased.The levels of NE(P〈0.05),DA(P〈0.01)and 5-HT(P〈0.01)were restored.Conclusion The novel object recognition test result demonstrated that Naosuikang decoction could improve the episodic memory ability in VCI rats and this may effect through regulating oxidative stress and the levels of monoamine neurotransmitters.
