摘要
目的:探讨原发性干燥综合征(p SS)患者外周血环状RNA(circRNA)的差异性表达及其与发病机制的相关性。方法:首先对4例p SS患者及4例健康对照者外周血的circRNA采用circRNA芯片进行筛查,然后使用Gene Spring软件对实验数据进行归一化及后续处理,通过Fold Change和P值筛选,对比两组间circRNA表达的差异性并对差异性进行生物学分析。结果:circRNA芯片筛查结果提示:病例组和对照组之间有437个circRNA差异具有统计学意义,其中上调circRNA 365个、下调circRNA 72个。通过对这437个circRNA相关基因进行基因本体论(GO)和通路(Pathway)分析,结果显示:GO分析表明这些基因主要参与生物调节、细胞组分、代谢及酶活性的调节等,KEGG Pathway分析表明这些基因主要参与氨基酸的降解与合成。并对具有差异性表达的circRNA靶miRNA进行预测,预测结果发现其中有1个circRNA(hsa-circRNA12807-10)中的1个miRNA(hsa-miR-181a-5p)在有关文献中得到了验证。结论:p SS患者外周血中具有多个差异性表达circRNA,同时这些差异性表达的circRNA可能与p SS的发病机制有关。
Objective :To investigate the role of circRNA in the pathogenesis of primary Sjogren's syndrome(pSS). Methods :The circcNA microarray was performed to screen the eircRNA in the peripheral blood from 4 patients with pSS and another 4 healthy controls. The experimental data were normalized and processed hy Gerie Spring software. The Fold Change and P-value were used to compare the eireRNA expression between groups, and the expression difference was analyzed hy hioinformaties. Results : Mieroarray indicated significant differenee between the case group and control group in a total of 437 eireRNAs,in which 365 were up-regulated, and 72 down-regulated. Analysis hy gene ontology(GO) and KEGG pathway showed that these genes were mainly involved in the regulation of biological regulation,cell composition,metabolism and enzyme activity as well as in the degradation and synthesis of amino acids. Differentially expressed circRN A target mibtN As were further predicted, and one of miRN As ( hsa-miR- 181 a-Sp) targeted hy a eircbtNA ( hsa- circRNA 12807-10) was confirmed in the literature. Conclusion:Several eireRNAs are differentially expressed in the peripheral blood of patients with pSS, suggesting that these differentially expressed genes may he involved in the pathogenesis of pSS.
