摘要
目的:通过观察Ⅰ型单纯疱疹病毒(HSV-1)对牙龈成纤维细胞炎症因子分泌情况的影响,探究HSV-1与牙龈炎症之间的相关性。方法:体外培养人牙龈成纤维细胞,分别用1×10~4PFU/m L、1×10~5PFU/m L、1×10~6PFU/m L 3种浓度的HSV-1病毒加以刺激,48 h、96 h后分别收集各组上清液进行ELISA检测,观察白细胞介素-6(IL-6)的表达情况; 96 h后提取细胞RNA,real-time PCR检测细胞IL-6基因的表达情况。结果:3组实验组的上清液中均有IL-6的表达,且随着刺激浓度的加大、刺激时间的延长,表达量增加; real-time PCR结果显示与对照组相比随着浓度的加大IL-6 mRNA表达水平增加,但1×10~5PFU/m L、1×10~6PFU/m L浓度组之间差异无统计学意义(P>0.05)。结论:Ⅰ型单纯疱疹病毒(HSV-1)能增加炎症因子IL-6的表达,并对人牙龈成纤维细胞产生影响,可能会增加牙龈炎症的易感性。
Objective: To investigate the effect of herpes simplex virus type 1 (IISV-1) on the secretion of inflammatory- foctors in human gingival fibro- blasts ,and correlation of IISV-1 with gingival inflammation. Methods:lluman gingival fibroblasts were cultured in vitro,and challenged with IISV-1 in dose of 1×10^-4PFU/mL,1×10^5PFU/mL,1×10^6PFU/mL3,respectively. The supernatant was harvested from each group after 48 h and 96 h of incuba- tion,and subjected to detection of interleukin-6(IL-6) expression using ELISA. Real-time PCR was performed to detect the cell RNA and IL-6 expression in the supernatant obtained at 96 h. Results : IL-6 was expressed in the supernatant of the three experimental groups, and up-regulated with added dose and prolonged challenge. Real-time PCR indicated that IL-6 mRNA was also increased with I ISV-1 dose as compared with the control group,yet the difi^renee was insignificant between dosage groups of 1 × 10^5 PFL/mL and 1 × 10^6 PFL/mL ( P〉 0.05).Conclusion:11SV-1 can increase IL-6 expression and change the biological activity of human gingival fibroblasts ,which may lead to increased susceptibility to gingival inflammation .
