枳朴中空栓对术后肠麻痹大鼠肠道NOS神经元的影响

摘要 目的:探讨枳朴中空栓在防治术后肠麻痹方面的作用机制。方法:选用雌性大鼠,切除子宫建立术后肠麻痹模型,空白组与模型对照组大鼠均给予空白中空栓,一次1粒,2次/天;枳朴中空栓组给予枳朴中空栓,一次1粒,2次/天。连续给药2天后脱臼处死,制作回肠、结肠肌间神经丛全层标本,通过双重免疫荧光染色法观察回肠、结肠肌间神经丛NOS神经元的变化。结果:模型对照组较空白组NOS神经元所占比例明显增高(P <0.01);枳朴中空栓组较模型对照组NOS神经元阳性表达降低(P <0.05)。结论:枳朴中空栓防治术后肠麻痹机制在于有效降低肌间神经丛NOS神经元的阳性表达,减少NO的含量,从而增加肠道平滑肌的收缩。 Objective To explore efects of Zhipo Zhongkong Shuan (枳朴中空栓 ) on enteroparalysis. Methods: The female rats were used for establishing rat models of postoperative ileus by removing uterus. The blank group and model control group were given Zhongkong Shuan ( 中空栓 ), 1 grain per time, 2 times a day. The Zhipo Zhongkong Shuan group was given Zhipo Zhongkong Shuan, 1 grain per time, 2 times a day. After taking the medicine for 2 days, then dislocated executed, the whole layer of the ileum and colonic intermuscular plexus was made, then the change of the nitric oxide synthase neurons in ileal and colonic myenteric plexus was detected by the dual immunofuorescent staining. Results: The proportion of NOS neurons in the model control group was signifcantly higher than that in the blank group (P〈0.01). The positive expression of the NOS neurons in the Zhipo Zhongkong Shuan group was lower than model control group (P〈0.05). Conclusion: Zhipo Zhongkong Shuan could efectively reduce positive expression of the NOS neurons in myenteric plexus of intestinal tract in rats with enteroparalysis and NO production, and increase contraction in the intestinal smooth muscle.
