摘要
目的:研究丹红注射液对脑梗死脑缺血再灌注大鼠脑水肿的影响。方法:采用大鼠大脑中动脉闭塞法制作局灶性脑缺血再灌注模型。应用干-湿比重法测定脑含水量,免疫组化SABC法测定水孔蛋白4(AQP4)的表达,化学比色法测定脑组织中的Na-K-ATP酶活性。结果:模型组AQP4蛋白表达升高;予丹红注射液的治疗组AQP4蛋白的表达下降;模型组脑组织匀浆Na-K-ATP酶活性明显低于正常对照组,予丹红注射液的治疗组则明显高于模型组。结论:脑组织缺血区AQP4蛋白表达的上调和Na-K-ATP酶活性降低可能与脑水肿的发生发展有一定的关系,丹红注射液有抑制脑水肿发生的作用。
Objective: To study the efcacy of the Danhong injection ( 丹红注射液 ) on cerebral edema after cerebral ischemia reperfusion in rats. Methods: The model of focal cerebral ischemia reperfusion was made by middle cerebral artery occlusion method. The content of brain was determined by the dry-wet specifc gravity method. The expression of water pore protein 4 (AQP4) was determined by immunohistochemical SABC method. The activity of NA and K-ATP enzyme in brain tissue was determined by chemical colorimetric method. Results: The expression of AQP4 protein in the model group was elevated. The expression of AQP4 protein in the treatment group was decreased. The activity of Na-K-ATP in the model group was signifcantly lower than that in the control group; and that in the treatment group was signifcantly higher than the model group. Conclusion: The increase of expression of AQP4 protein and the decrease of activity of NA-K-ATP enzyme in cerebral ischemic region may be related to the occurrence and development of brain edema. The Danhong injection can inhibit the occurrence of brain edema.
