摘要
线粒体酶对线粒体正常功能的行使具有重要作用,重金属对线粒体酶的结构和功能都具有显著的影响,会使其发生不可逆的损伤,从而影响线粒体的正常功能。以小鼠肝脏线粒体酶为研究对象,探究50、100、200、300、400和500 mg/kg等不同浓度铅胁迫下5种线粒体酶活性的变化。实验结果显示,随着Pb2+浓度的增加,小鼠肝脏线粒体SDH、NADH、ATP、GST和GSH-PX 5种酶的活性都呈现下降趋势,当Pb2+浓度为500 mg/kg时,酶活性下降至最低点,较对照组差异极显著(P <0. 01)。统计学分析表明,各实验组酶活性与铅离子浓度呈显著负相关性。
Mitochondrial enzymes play important role for cell, but the morphological characteristics, structure and function of enzymes would be changed significantly stressed by heavy metals, and also will be irreversible damaged. This study takes the liver mitochondria of mice as the subject of study under the 50mg/kg, 100mg/kg, 200mg/kg, 300mg/kg, 400mg/kg and 500mg/kg of different concentrations of Pb^2+ stress to explore the five mitochondrial enzyme activities. The results showed that SDH, NADH, ATP, GST and GSH-PXG activity decreased as Pb^2+ dosage increased. And all reached the bottom after 500mg/kg, which had significant differences compared with controls ( P 〈0.01). Statistical analysis showed that SDH, NADH, ATP, GST and GSH-PX activity had a significant negative correlation with Pb^2+ concentration.
