摘要
目的探讨山柰挥发油胃内滞留型给药系统对胃肿瘤治疗的作用机理。方法常规培养胃癌细胞SGC-7901,实验分组为对照组(C组)、山柰挥发油低剂量处理组(20μg/ml)、山柰挥发油高剂量处理组(60μg/ml),利用MTT检测各组细胞增殖,流式检测细胞凋亡和细胞周期,western blot和PCR检测细胞中COX-2和AFP的表达情况。结果与对照组相比,20μg/ml低剂量药物处理组和60μg/ml高剂量药物处理组细胞增殖率明显降低,凋亡率随着药物浓度的增加也明显增高。加入药物处理后,细胞周期中,处于S期的细胞量变少,G1期细胞增加,表明药物可以抑制胃癌细胞DNA的合成。与对照组相比,低剂量药物处理组和高剂量药物处理组的COX-2和AFP表达明显降低。结论山柰挥发油可以影响胃癌细胞的增殖、凋亡和周期,同时也能够影响细胞内蛋白和相关基因的表达。
Objective To explore the mechanism of gastric retention drug system of hawthorn volatile oil on gastric cancer treatment. Methods The gastric cancer cells SGC-7901 were routinely cultured.They were divided into the control group (C group),Hawthorn volatile oil low-dose treatment group (20 μg/ml),and Hawthorn volatile oil high-dose treatment group (60 μg/ml),then use MTT to detect the proliferation of each group,flow cytometry was used to detect cell cycle and apoptosis,and expression of COX-2 and AFP in cells was detected by western blot and PCR. Results Compared with the control group,the proliferation rate of the 20 μg/ml low-dose drug treatment group and the 60 μg/ml high-dose drug treatment group significantly decreased,and the apoptotic rate also increased significantly with the increase of the drug concentration.After treatment with drugs,the amount of cells in the S phase decreased and the number of cells in the G 1 phase increased during the cell cycle,indicated that the drug can inhibit DNA synthesis in gastric cancer cells.Compared with the control group,the expression of COX-2 and AFP significantly decreased in the low-dose drug treatment group and the high-dose drug treatment group. Conclusion Hawthorn volatile oil can affect the proliferation,apoptosis and cycle of gastric cancer cells,and can also affect the expression of intracellular proteins and related genes.
