摘要
为了测定梵净山野生八角莲中槲皮素和山奈酚的含量,采用高效液相色谱法,在色谱柱为Inert Sustain C18柱(250×4.6mm,5μm),乙腈-0.4%磷酸(体积比35∶65)作为流动相,检测波长362nm,柱温25℃,流速0.7m L/min的最佳色谱条件下进行研究,结果表明:八角莲的干燥根茎中含有槲皮素和山奈酚两种黄酮类化合物,根据槲皮素和山奈酚的标准曲线得出,槲皮素在0.0075~0.0375 mg/m L范围内线性关系良好(R^2=0.9999,n=5);山奈酚在0.007~0.035 mg/m L范围内线性关系良好(R^2=0.9998,n=5);槲皮素的平均回收率为100.90%,相对标准偏差(RSD)为2.49%,山奈酚的平均回收率为101.07%,相对标准偏差(RSD)为3.81%,并对不同产地的含量进行比较,结果表明:产地不同,其含量有一定差异,梵净山的含量较高,槲皮素为5.92%,山奈酚为1.95%。由此可知:此方法简单,快速,精确,重复性好,可为本地八角莲药材质量控制提供一种有效方法。
We studied a method of high performance liquid chromatography to determine Dysosma quercetin and kaempferol contents. Using high performance liquid chromatography under opfimunl chromatographic conditions:The colunm was a colunm of InertSustainC18 (250 ×4.6mm,5μm) and aeetonitrile -0.4% phosphoric acid (35:65 by volume) as the mobile phase. The detection wavelength was 362nm, eolunm temperature was 25 ℃ and flow rate was 0.7mL/nfin. The results showed that Quereetin and Kaempferol were contained in the dried Dysosma Versipellis, according to the standard curve of Quereetin and Kaempferol, Quercetin had a good linearity in the range of 0.0075 - 0. 0375 mg/mL( R2 = 0.9999, n = 5 ) ; The linear range of Kaempferol was in the range of 0.007 -0. 035 mg / mL (R2 = 0. 9998, n = 5 ) ;The average recoveries of Quercetin were 100. 90% and the relative standard deviations (RSD) were 2.49%. The average recoveries of Kaempferol were 101.07% and the relative standard deviations (RSD) were 3.81%. The results showed that there was a certain difference in the content, the content of Sanskrit mountain was higher, the quereetin was 5.92% , and the mountain neophenol was 1.95%. It can be seen:This method is simple, rapid, accurate, reproducible, and can provide an effective method for the Dysosma medicine quality control.
