DDX60在结直肠癌中表达特点的研究

The Expression Profile of DEXD/H box helicase 60 in Colorectal Cancer

目的:探讨DDX60 (DEXD/H box helicase 60)在小鼠结直肠癌(Colorectal cancer,CRC)形成过程中及在临床CRC样本中的表达及意义。方法:通过致癌剂氧化偶氮甲烷(Azoxymethane,AOM)和致炎剂葡聚糖硫酸钠(Dextran sulfate sodium,DSS)的协同作用,构建经典的炎症相关性CRC模型。在CRC形成后的第80天和形成早期的第14天收集结肠组织,通过qRT-PCR和Western-blot检测DDX60基因及蛋白的表达。同时,收集临床CRC组织及相应癌旁组织样本,通过qRT-PCR和Western-blot检测临床CRC样本中DDX60基因及蛋白的表达。结果:与对照组相比,DDX60在小鼠CRC模型及人CRC组织中的表达均呈显著性降低(P <0. 05)。结论:DDX60的低表达可能是CRC的促因之一,DDX60可能具有抑制CRC的发生发展和治疗CRC的潜能。

Objective： This study is to investigate the expression of DDX60 in colon tissues from mice under the process of CRC formation as well as from clinical CRC samples. Methods： The classical inflammation-related CRC model was used, which constructed by the synergistic effect of carcinogen Azoxymethane （AOM） and Dextran sulfate sodium （DSS）. Mouse colon tissues on day 80 after tumor formation and day 14 at the early time point were collected. Through qRT-PCR and Western-blot, the expression of DDX60 gene and protein was measured. Meanwhile, mRNA and protein expression of DDX60 in human CRC samples were also determined by qRT-PCR and Western-blot. Results： In comparison with the control group, the expression of DDX60 in mouse colon tissues and human CRC tissues significantly decreased （P〈0.05）. Conclusion： The decreased expression of DDX60 in colorectal tissues may be involved in process of CRC formation,and DDX60 may have suppressive effect and therapeutic value for CRC.
