摘要 目的探讨葛根素对宫颈癌细胞迁移及侵袭的影响。方法分别采用浓度为0(对照组)、12.5、25、50μmol/L的葛根素作用Hela细胞48 h。CCK-8法检测细胞活力,transwell法检测细胞迁移及侵袭能力,western blot检测基质金属蛋白酶2(MMP2)、MMP9、E-cadherin、Vimentin及转化生长因子-β1(TGF-β1)/Smad信号通路相关蛋白表达。结果与对照组比较,12.5、25、50μmol/L葛根素作用48 h后可显著降低Hela细胞活力[(0.571+0.057)vs(0.462+0.045)、(0.394+0.039)、(0.323+0.033)],迁移能力[(245.68±24.56)vs(189.56±18.95)、(108.28±10.84)、(53.59±5.35)]及侵袭能力[(109.67±10.97)vs(78.56±7.85)、(43.46±4.34)、(22.39±2.23)],且上调E-cadherin表达,下调MMP2、MMP9、Vimentin、TGF-β1、p-Smad2及p-Smad3表达,P均<0.01。结论葛根素可显著抑制宫颈癌细胞的迁移及侵袭能力。 Objective To explore effect of puerarin on migration and invasion in cervical cancer cells.Methods Hela cell was cultured with 0( control),12.5,25,50 μmol/L dose of puerarin for 48 h. Cell viability was measured by CCK-8 assay.Cell migration and invasion ability was detected by transwell assay. The expression of matrix metalloproteinase 2( MMP2),MMP9,E-cadherin,Vimentin,transforming growth factor-β1( TGF-β1)/Smad signal pathway related protein was measured by western blot.Results Compared with control,12.5,25,50 μmol/L dose of puerarin reduced cell viability [( 0.571+0.057) vs( 0.462+0.045),( 0.394+0.039),( 0.323+0.033) ],migration [( 245.68±24.56) vs( 189.56±18.95),( 108.28±10.84),( 53.59±5.35) ]and invasion ability [( 109.67±10.97) vs( 78.56±7.85),( 43.46±4.34),( 22.39±2.23) ],up-regulated the expression of E-cadherin,down-regulated the expression of MMP2,MMP9,Vimentin,TGF-β1,p-Smad2 and p-Smad3.Conclusion Puerarin could inhibit cervical cell migration and invasion.
