丙泊酚下调ERK信号通路对乳腺癌细胞增殖和转移能力的影响 被引量:8

摘要 目的研究丙泊酚对人乳腺癌细胞增殖转移能力的影响及潜在作用机制。方法 MTT检测不同时间(0、12、24、48、72 h)和不同浓度(0、12.5、25、50、100μmol/L)丙泊酚对人乳腺癌细胞MDA-MB-231增殖的影响;Transwell实验检测100μmol/L的丙泊酚对细胞迁移侵袭能力的影响;Western blot检测100μmol/L的丙泊酚分别作用0、12、24、48、72 h后细胞中MMP9、Cyclin D1、ERK蛋白表达变化。结果丙泊酚可呈时间-剂量依赖性地抑制MDA-MB-231细胞增殖;丙泊酚(100μmol/L)可显著抑制细胞迁移侵袭能力;丙泊酚可呈时间依赖性地抑制细胞中MMP9、Cyclin D1、p-ERK蛋白表达。结论丙泊酚能够抑制人乳腺癌细胞增殖转移能力,其机制可能与下调ERK信号有关。 Objective To investigate the effect of propofol on the proliferation and metastasis of human breast cancer cells and its potential mechanism. Methods The effects of propofol on the proliferation of human breast cancer cells MDA-MB-231 at different time points( 0,12,24,48,72 h) and different concentrations( 0,12.5,25,50,100 μmol/L) were detected by MTT. The effect of 100 μmol/L propofol on cell migration and invasion was examined by Transwell assay. Western blot was used to detect the expressions of MMP9,Cyclin D1 and ERK in cells treated with 100 μmol/L propofol for 0,12,24,48 and 72 h,respectively.Results Propofol inhibited the proliferation of MDA-MB-231 cells in a time-and dose-dependent manner. Propofol( 100 μmol/L)significantly inhibited cell migration and invasion. Propofol could inhibit the expressions of MMP9,Cyclin D1 and p-ERK protein expression.Conclusion Propofol can inhibit the proliferation and metastasis of human breast cancer cells,and the mechanism might be related to down-regulation of ERK signaling.
