低氧环境下SDF-1对间充质干细胞迁移影响的初探 被引量:1

摘要 目的探讨低氧环境下基质细胞衍生因子(SDF-1)促进间充质干细胞的迁移影响。方法 2017年3月—2018年3月期间利用三气培养箱建立低氧微环境,加入120 ng/mL浓度SDF-1间充质干细胞为实验组,在低氧环境下培养的间充质干细胞为低氧对照组,在正常环境下培养的间充质干细胞为正常对照组;利用Transwell细胞迁移实验检测各组细胞迁移;利用MTT实验检测每组间充质干细胞的增殖抑制率。结果 Transwell细胞迁移实验表明细胞迁移率实验组(43.5±8.4)×10~3与正常对照组(26.7±5.6)×10~3比较差异有统计学意义(P<0.01),低氧对照组(37.5±7.2)×10~3与正常对照组(26.7±5.6)×10~3相比较差异有统计学意义(P<0.05);在低氧环境下,与低氧对照组相比较,实验组间充质干细胞的细胞活性明显升高(P<0.01);同时低氧对照组与正常对照组相比间充质干细胞的细胞活性也明显升高(P<0.01)。结论在低氧环境下加入趋化因子SDF-1可有效的趋化间充质干细胞,在低氧环境下加入趋化因子度SDF-1因子对间充质干细胞活性增强。 Objective To investigate the effect of stromal cell-derived factor (SDF-1) on the migration of mesenchymal stem cells under hypoxic conditions. Methods From March 2017 to March 2018, three gas incubators were used to establish a hypoxic microenvironment. SDF-1 mesenchymal stem cells with a concentration of 120 ng/mL were used as the experimental group. The mesenchymal stem cells cultured under hypoxic conditions were in the hypoxic control group, the mesenchymal stem cells cultured under normal environment were in normal control group; Transwell cell migration assay was used to detect cell migration in each group; MTT assay was used to detect the proliferation inhibition rate of mesenchymal stem cells in each group. Results Transwell cell migration assay showed that the cell migration rate in the experimental group (43.5±8.4)×10^3 was significantly different from the normal control group (26.7±5.6)×10^3 (P〈0.01), and the hypoxic control group (37.5±7.2)×10^3. Compared with the normal control group (26.7±5.6)×10^3, there was a significant difference (P〈0.05). Under hypoxic conditions, compared with the hypoxic control group, the cell activity of the mesenchymal stem cells in the experimental group was significantly increased (P〈0.01); At the same time, the cell activity of mesenchymal stem cells in the hypoxic control group was also significantly higher than that in the normal control group (P〈0.01). Conclusion The addition of chemokine SDF-1 in hypoxia can effectively metastasize mesenchymal stem cells. The activity of mesenchymal stem cells is enhanced by the addition of chemokine factor SDF-1 in hypoxia.
