摘要 目的:用RP-HPLC联合紫外检测器同时测定解毒消炎茶中绿原酸等9种活性成分含量。方法:采用Agilent C18(250. 0 mm×4. 6 mm,5μm)色谱柱,以乙腈(A)-0. 4%磷酸水(B)系统作为流动相,梯度洗脱(0~25 min,5%~22%A;25~31 min,22%~25%A;31~45 min,25%~45%A;45~60 min,45%~55%A;60~80 min,55%~100%A);柱温为30℃,检测波长为320 nm,流速为1. 0 m L/min。结果:绿原酸、连翘酯苷A、栀子苷、橙皮苷、连翘苷、芦荟大黄素、大黄酸、大黄素、大黄酚分别在138. 00~5520. 00 ng,30. 56~1222. 40 ng,84. 80~3392. 00 ng,108. 48~4 339. 20 ng,20. 72~828. 80 ng,20. 88~835. 20 ng,25. 76~1 030. 40 ng,33. 92~1 356. 80 ng,60. 24~2 409. 60ng范围内呈良线性关系,相关系数均大于0. 999;平均回收率均在98%~102%范围内。结论:RP-HPLC联合紫外检测器同时测定解毒消炎茶中绿原酸等9种不同极性成分灵敏、准确,分离效果较好,可为解毒消炎茶质量标准评价提供依据。 Objective:To determine the content of Chlorogenic acid and other eight active ingredients in JieduXiaoyan tea by RP-HPLC combined UV detector with simultaneous determination.Methods:The chromatographic column was Agilent C18 (250 mm×4.6 mm,5 microns) column,with acetonitrile (A) -0.4% phosphoric acid water system (B) as mobile phase,gradient elution 0~25 min,5% A~22% A;25~31 min,22%~25% A;31~45 min,25%~45% A;45~60 min,45%~55% A;60~80 min,55%~100% A).Column temperature was 30 ℃,with detection wavelength of 320 nm and the flow rate of 1.0 mL/min.Results:Chlorogenic acid,Forsythoside A,Geniposide,hesperidin,Phillyrin,aloeemodin,rhein,emodin,chrysophanol had good linear relationship within the range of 138.00~5 520.00 ng,30.56~1 222.40 ng,84.80~3 392.00 ng, 108.48~4 339.20 ng,20.72~828.80 ng,20.88~835.20 ng,25.76~1 030.40 ng,33.92~1 356.80 ng,60.24~ 2 409.60 ng,respectively,and the correlation coefficient were greater than 0.999.The average recovery rate were all in the 98%~102% range.Conclusion:The method on Simultaneous determination of content of Chlorogenic acid and other eight eight different polarity ingredients in JieduXiaoyan tea by RP-HPLC combined UV detector have high sensitivity,high accuracy and ideal separation effect,can be used as an jieduxiaoyan tea quality evaluation.
