氧化苦参碱-黄芪多糖中药复方对雏鸡血清中抗氧化指标的影响 被引量:6

Effects of oxymatrine-astragalus polysaccharide compound on the antioxidant indexes in the serum of chickens

摘要 为研究氧化苦参碱-黄芪多糖中药复方在鸡生产中的应用效果,测定了不同剂量氧化苦参碱-黄芪多糖中药复方(OAP)对雏鸡血清抗氧化性能的影响。选择120羽14日龄非免疫健康罗曼公雏鸡,随机均分为4组,Ⅰ、Ⅱ、Ⅲ组分别口服低剂量(1 mg/mL)、中剂量(2 mg/mL)和高剂量(4 mg/mL)的氧化苦参碱-黄芪多糖中药复方,每羽1 mL,Ⅳ组为口服等量生理盐水的空白对照组,分别于试验开始后的第7、14、21和28天时采血,测定血清中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、谷胱甘肽还原酶(GR)和丙二醛(MDA)活性。结果:雏鸡血清SOD含量在第28天时Ⅰ组与Ⅳ组差异极显著(P〈0.01),提高17.81%,Ⅱ组在试验第7、14、21和28天与Ⅳ组雏鸡血清SOD含量均出现差异极显著(P〈0.01),分别提高16.23%、35.96%、25.08%和30.53%;Ⅱ组在试验全期与Ⅳ组雏鸡血清GSH-Px含量差异极显著(P〈0.01),分别提高25.08%、19.48%、25.50%和15.91%;雏鸡血清GR在第14天时,Ⅰ、Ⅱ、Ⅲ组与对照组差异显著(P〈0.01),分别提高了30.09%、54.03%、30.02%,第21天时雏鸡血清GR差异显著(P〈0.01),分别提高了18.78%、39.28%和18.96%,第28天时雏鸡血清GR差异显著(P〈0.01),分别提高了26.41%、48.24%和25.25%;Ⅰ、Ⅱ、Ⅲ组在整个试验期间雏鸡血清中MDA含量都有不同程度降低,Ⅰ、Ⅲ组在试验全期与Ⅳ组雏鸡血清中MDA含量出现显著差异(P〈0.05),Ⅱ组在试验全期与对照组雏鸡血清中MDA含量出现极显著差异(P〈0.01)。综上,3种不同剂量氧化苦参碱-黄芪多糖中药复方均对机体抗氧化能力能显著改善,以Ⅱ组中剂量(2 mg/mL)最为适宜。 To study the effect of oxymatrine-astragalus polysaccharide(OMT-APS) on antioxidant enzyme activity in chickens and provide theoretical basis for application of OMT-APS to production of chickens,120 healthy 14-day old male chicks with no immunity were selected and randomly divided into four groups(n = 30). The chickens of groups Ⅰ,Ⅱ and Ⅲ were treated with a low dose(1 mg/mL),a medium dose(2 mg/mL),and a high dose(4 mg/mL) of OMT-APS,respectively. Group Ⅳ(The control group) was given the same amount of saline. Blood samples were obtained at day 7,day14,day 21 and day 28 of the trial in order to determine the activities of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),glutathione reductase(GR) and malondialdehyde(MDA) in serum. The experimental results were: The difference in SOD in serum between groups I and Ⅳ was significantly(P0. 01) increased at day 28 by 17. 81%. This difference between groups Ⅱ and Ⅳ were significantly(P0. 01) increased at days 7,14,21 and 28 by 16. 23%,35. 96%,25. 08%、30. 53%,respectively. The difference in GSH-Px in serum between groups Ⅱ and Ⅳ was significantly(P〈0. 01) increased at the four time points by25. 08%,19. 48%,25. 50% and 15. 91%,respectively. The difference in GR in serum between groups Ⅰ,Ⅱ,Ⅲ,and Ⅳ was significantly(P〈0. 01) increased at day 14 by 30. 09%,54. 03% and 30. 02%,respectively; at day 21 by 18. 78%,39. 28% and 18. 96%,respectively; at day 28 by 26. 41%,48. 24% and 25. 25%,respectively. The MDA in serum of groups Ⅰ,Ⅱ and Ⅲ were reduced to different degrees during the whole experiment,with a significant difference between groups Ⅰ,Ⅲ and Ⅳ(P〈0. 05) and between groups Ⅱ and Ⅳ(P〈0. 01). In summary,the three different doses of oxymatrine and astragalus polysaccharide compound significantly improved the antioxidant activity in chickens,with the medium dose being the most effective in chickens.
