骨炎方抗家兔慢性骨髓炎的实验研究 被引量:3

Experimental Study of Guyan Prescription on Chronic Osteomyelitis in Rabbits

摘要 目的:观察骨炎方对家兔慢性骨髓炎的作用。方法:家兔右胫骨骨髓腔内注射金黄色葡萄球菌建立慢性骨髓炎模型,分为正常对照组、模型组、庆大霉素组(60mg/kg)、骨炎方高剂量组(生药3.6g/kg)、骨炎方低剂量组(生药1.8g/kg),每组10只,灌胃给药,术后第6周对右胫骨行X-线检查并做Laurence评分;术后第2,4及6周采血测定白细胞计数、血清溶菌酶、血清TNF-α和IL-6含量。结果:模型组家兔均有典型骨髓炎表现,骨炎方高、低剂量组对家兔骨髓炎症状有显著缓解,Laurence评分与模型组比较差异均有统计学意义(P值分别为0.000,0.001);与模型组比较,骨炎方高剂量组白细胞计数在第2,4及6周均有明显降低,差异有统计学意义(P值分别为0.015,0.012,0.001),低剂量组白细胞计数在第2,4及6周均有明显降低,差异有统计学意义(P值分别为0.013,0.008,0.036),高剂量组血清溶菌酶含量在第2,4及6周均有明显升高,差异有统计学意义(P值分别为0.033,0.002,0.003),低剂量组血清溶菌酶含量在第2,4及6周亦有明显升高,差异有统计学意义(P值分别为0.048,0.028,0.024),高剂量组血清TNF-α含量在第2,4及6周均有明显降低,差异有统计学意义(P值分别为0.031,0.003,0.000),低剂量组血清TNF-α含量在第2,4及6周均有明显降低,差异有统计学意义(P值分别为0.032,0.008,0.000),高剂量组血清IL-6含量在第2,4及6周均有明显降低,差异有统计学意义(P值分别为0.004,0.000,0.000);低剂量组血清IL-6含量在第2,4及6周均有明显降低,差异有统计学意义(P值分别为0.009,0.000,0.000).结论:骨炎方对家兔慢性骨髓炎的防护作用可能与抑制致病菌、提高机体免疫力、下调炎性因子等途径有关。 Objective:To investigate the effects of Guyan prescription(GP)against chronic osteomyelitis in rabbits.Methods:Chronic osteomyelitis model of rabbits were prepared by intra-shinbone marrow injection of Staphylococcus aureus.Fifty rabbits were divided randomly into five groups(n=10),i.e.normal,model,gentamicin(60mg/kg),GP high and low dose groups(3.6and 1.8g/kg,respectively).Drugs were orally administrated after the operation.X-ray examination of the right shinbone at the sixth week after operation was done and Laurence score were calculated.At the second,fourth and sixth week after operation,blood was collected to determine the leukocyte count,serum lysozyme,serum TNF-αand IL-6levels.Results:Rabbits in the model group all showed typical osteomyelitis.The high-and low-dose groups of GP significantly alleviated the symptoms of osteomyelitis in the rabbits,and the Laurence scores were statistically significant compared with the model group(Pvalues were 0.000,0.001,respectively).Compared to model group,leucocyte count of the high-dose GP group had a significant decrease at the second,fourth and sixth weeks(P=0.015,0.012,0.001),so did the low-dose GP group(P=0.013,0.008and 0.036,respectively).Serum lysozyme levels increased significantly both in the high-dose group(Pvalues were 0.033,0.002,and 0.003,respectively)and low dose group(P=0.048,0.028and0.024,respectively)at the 2nd,4th,and 6th weeks.The levels of serum TNF-αboth in the high-dose group and in the low-dose group(P=0.032,0.008 and 0.000,respectively)decreased significantly at the 2nd,4th,and 6th week,and the difference was statistically significant(P values were 0.031,0.003,and 0.000,respectively).The serum IL-6levels both in the high-dose group(P values were 0.004,0.000,and 0.000,respectively)and in the low-dose group(P values were0.009,0.000,0.000 respectively)were significantly lower at the second,fourth,and sixth weeks.Conclusion:The protective effect of GP on osteomyelitis seems to be associated with inhibition of pathogenic bacteria,improvement of immune function and down regulation of inflammatory cytokines.
